摘  要:选用39个虾夷扇贝Patinopecten yessoensis多态性微卫星分子标记对大连獐子岛海域虾夷扇贝一个家系群体(以獐子岛海域底播虾夷扇贝和日本产野生虾夷扇贝为父母本交配产生的F1代)的有效等位基因数(Ne)、观测杂合度(Ho)、期望杂合度(He)、多态信息含量(PIC)等进行了检测,以卡方检验估计群体Hardy—Weinberg平衡。结果表明:在39个基因座位中共检测到105个等位基因,每个座位检测到的等位基因数为2—4个不等,等位基因片段大小为95~440bp,平均等位基因数为2.70个,平均有效等位基因数为2.40个,观测杂合度平均值0.6280,期望杂合度平均值为0.5240,平均多态信息含量为0.4556,经卡方检验(P〈0.01)值显示多于半数的位点都发生了偏离。采用SAS8.2软件中的一般线性模型(GLM)对39个微卫星标记与虾夷扇贝群体的生长性状进行连锁显著性检验。结果表明:在39个微卫星座位中,HLJX202位点与体重、壳长、壳高、壳宽都显著相关,HLJX109、HLJX196位点与体重、壳长、壳高显著相关,HLJX183位点与壳长、壳高显著相关,而HLJX128和HLJX236位点分别只与壳宽和体重显著连锁。The genetic structure of one breeding population of Japanese scallop (Patinopecten yessoensis) was analyzed using thirty - nine polymorphic microsatellite markers. The effective number of alleles ( Ne ), observed het- erozygosity ( Ho ) , expected heterozygosity ( He ) and polymorphism information content(PIC) were all determined. The Hardy - Weinberg equilibrium was checked by chi - square test. For each locus, 2 - 4 alleles were detected, with a total of 105 alleles for 39 loci. The average effective number of alleles and the mean polymorphism information content were 2.40 and 0.4556, respectively. Observed heterozygosity and expected heterozygosity were 0. 6280 and 0.5240, respectively. The probability value of chi - square test showed that more than half of the thirty - nine loci had significantly deviated from Hardy - Weinberg equilibrium( P 〈 0.01 ). The SAS 8.2 was used to analyze the effect of these 39 microsatellites on body weight, shell length, width and height. Results indicated that HLJX109, and HLJX196 had a significant impact on body weight and shell length ; HLJX128 and HLJX236 had a significant impact on shell width and body weight,respectively; HLJX183 had a significant on shell length and body weight. Several significantl diversity loci were also analyzed by the multiple comparisons of Duncan.
