摘  要:自人胚颅骨分离出的成骨细胞在含50mg/LVitC和10mmol/Lβ-甘油磷酸钠的M199+20%新生牛血清培养液内逐渐聚集、重叠而呈集落样生长,集落的中央有大量折光性强的颗粒沉积,经扫描电镜观察及电子探针分析,提示上述颗粒为钙盐沉积所致.表明体外培养的人胚成骨细胞可发生特异性钙化。Osteoblasts isolated from fetal human calvaria were cultured in the 199~20% FCS medium supplemented with 50 mg / L ascorbic acid and 10 mmol / L Na-Beta-glycerophosphate.Clumps and multilayer ofosteoblasts became evident with the increase of culture time.There were many granules which had strong dioptre in the center parts of the cell clumprs.The observation and analysis of the cell clumps by SEM and EPMA showed that the dioptric granules probably were mineral deposits or mineralized matrx.The results of the study indicate that fetal human osteoblast cultured in vitro elaborate the calcification,which further supportt the osteoblastic nature of the cells.
