摘  要:目的:研究虫草肾茶胶囊对培养在高糖条件下系膜细胞(HMC)增殖及细胞上清液中胶原纤维(Col-IV)的表达影响,探讨其在糖尿病肾病(DN)防治中的意义。方法:利用四甲基偶氮唑蓝(MTT)比色法动态观察各种处理因素的作用下各组HMC增殖情况;酶联免疫吸附试验(ELISA)测定经体外培养的人肾小球系膜细胞在各种处理因素的作用下上清液中IV型胶原的表达情况,并以福辛普利为对照。结果:高糖能明显刺激体外培养的HMC增殖,同时促进HMC分泌Col-IV(P<0.05或P<0.01)。而虫草肾茶胶囊能抑制HMC增殖及HMC分泌Col-IV,并呈一定量效关系(P<0.05或P<0.01),且某些方面明显优于福辛普利。结论:高糖可促进HMC增殖及Col-IV分泌,虫草肾茶胶囊能明显抑制高糖条件下HMC增殖及其分泌Col-IV,从而发挥预防肾小球纤维化及硬化的发生、发展,进而延缓DN的进展。Objective:To investigate the effect of Chongcao Shencha capsule on human mesangial cell(HMC) proliferation and the expression of collagen type IV(Col-IV) in vitro under high glucose.Methods: The thiazolyl blue tetrazolium bromide(MTT) colorimetric method was used to observe the human mesangial cell proliferation;Enzyme linked immunoadsordent assay(ELISA) was adopted to determine the expression of collagen type IV in cultured human mesangial cells,and establish the Fosinopril Sodium group.Results: In vitro cultured human mesangial cells were obviously proliferating under the stimulation of high glucose, which also promote Col IV secretion by human mesangial ceils ( P 〈 0.05 or P 〈0.01 ) ; but Chongcao Shencha capsule can inhibitc HMCs proliferating and secreting Col IV in a dose effect manner (P 〈0.05 or P 〈0.01 ), and in some respect the Chongeao Shencha capsule is more effective than the Fosinopril Sodium. Conclusion: High glucose can promote HMC proliferation and secretion of Col IV, but Chongcao Shencha capsule could inhibit it, which could be helpful for preventing of the progression of diabetic nephropathy.
