摘 要:目的:研究牡蛎钙补肾中药复方对骨形成蛋白-4(BMP-4)诱导成骨信号转导机制的调控作用。方法:采用切除大鼠双侧卵巢的方法复制绝经后骨质疏松症模型,应用牡蛎钙补肾中药复方进行防治,同时用骨疏康颗粒剂、牡蛎碳酸钙咀嚼片作为阳性对照,正常大鼠作为标准对照,模型大鼠作为模型空白对照。中药防治12周后进行取材及实验指标检测:用双能X线骨密度仪检测离体股骨骨密度;用W estern杂交法检测骨组织BMP-4和Sm ad5、6蛋白表达。结果:与模型空白组比较,牡蛎钙补肾中药复方各剂量组股骨骨密度显著升高(P<0.01);骨组织BMP-4、Sm ad5蛋白表达水平明显升高(P<0.01),Sm ad6蛋白表达水平明显降低(P<0.01)。结论:牡蛎钙补肾中药复方可明显上调BMP-4和Sm ad5蛋白表达,下调Sm ad6蛋白表达,这可能是其防治骨质疏松症的重要机制之一。Objective : To study the regulating effect of Chinese Herbal Compound Formula of Oyster calcium and invigorating kidney on bone morphogenetic protein(BMP) through inducing Signal Transduction Mechanism Change of BMP - 4. Methods : The models of postmenopausal osteoporosis were established through ovariectomy in rats. We treated the osteo- porosis rats with the Chinese Herbal Compound Formula of Oyster calcium and invigorating kidney. And we established the Gushukang Granule and Oyster Shell Carbonate Chewable Tablets as positive - control groups, the normal rats as standard -control group,and the common rats as blank control group. After 12 weeks of the treatment, the materials were obtained and the target was tested. The bone mineral density (BMD) of femoral bone in vitro was detected by dual -energy X -ray densitometer and the protein expression of BMP -4 and Smad 5,6 were determined by Western blotting. Results: Compared with the model rats, the BMD of femoral bone in vitro in the experimental drug groups( big dose,middle dose, small dose) increased obviously(P 〈 0.01 ). The protein expression of BMP - 4 and Smad5 increased obviously( P 〈 0.01 ), and the expression protein of Smad6 significantly decreased in the bone tissue ( P 〈 0.01 ). Conclusions : Chinese Herbal Compound Formula of Chinese Herbal Compound Formula of Oyster calcium and invigorating kidney could increase the protein expression of BMP -4 and Snaad5 and decrease the expression of Smad6 protein. This might be one of the mechanisms of prevention and treatment of Osteoporosis.
