摘  要:目的探讨丝裂原活化蛋白激酶MAPK在不同年龄小鼠卵母细胞中的表达差异及其与纺锤体染色体构象的关系。方法免疫细胞化学和激光共聚焦显微术检测6-8周和45-50周小鼠卵母细胞中纺锤体和染色体构象以及ERK1/2的表达差异。结果6-8周小鼠卵母细胞的纺锤体和染色体异常率为9%,明显低于45-50周小鼠的卵母细胞(29%P<0.025);6-8周小鼠卵母细胞中ERK1/2表达的荧光强度为78.21±13.34,明显高于45-50周小鼠的卵母细胞(61.45±15.67P<0.05)。结论卵母细胞纺锤体和染色体构象异常率与小鼠年龄存在明显的相关性,MAPK的表达降低可能是造成这种老龄卵母细胞纺锤体和染色体构象异常率增高的原因。Objective: To study the expression of mitogen activated protein kinase (MAPK) in mouse oocytes of different maternal age and its relationship to spindle and chromosome configuration. Methods: Spindle and chromosome configuration as well as expression of MAPK in mouse oocytes were detected by immunocytochemistry and confocal microscopy. Results: The abnormal spindle and chromosome configuration rate was 9% in oocytes of 6 - 8 week mouse, much lower than that of 45 - 50 week mouse ( P 〈 0. 025). The expression of ERK1/2 was (78.21 ± 13.34) in oocytes of 6 - 8 week mouse, much higher than that of 45 - 50 week mouse (61.45 ± 15.67, P 〈0. 05). Conclusion: Incidence of abnormal spindle and chromosome configuration in oocytes is related to maternal age, which may be due to the decrease expression of MAPK in aged oocytes.
