摘 要:目的:研究巴戟天正丁醇提物对乳鼠心肌细胞缺氧/复氧损伤(H/R)的防护作用。方法:采用培养的乳鼠心肌细胞建立缺氧/复氧损伤模型,实验分正常细胞培养组、缺氧/复氧损伤模型组、巴戟天醇提物小、中、大剂量组。观察心肌细胞形态并测定活性氧(ROS)、内皮素(ET-1)的含量变化。结果:巴戟天醇提物可明显改善缺氧/复氧损伤的心肌细胞形态及搏动程度;降低ROS、ET-1的含量。结论:巴戟天正丁醇提物对缺氧/复氧损伤的心肌细胞有防护作用;其防护作用可能与降低细胞液中ROS、ET-1的含量有关。OBJECTIVE To study the protective effect and its mechanisms of the n-butanol extract of morinda officinalis How(BEM) in cultured neonatal rat cardiomyoeytes by hypoxia/reoxygenation injury. METItODS The models of hypoxia/ reoxygenation were made with th first generation of cultured eardiomyocytes. Cultured eardiomyoeytes of neonatal rats were di vided into five groups: control group; hypoxia/reoxygenation (H/R) group; the groups of H/R + low dose of BEM(0. 038 g ·L^- 1 ), H/R + medium dose of BEM(0. 11g ·L^- 1) and H/R + high dose of BEM(0. 34 g ·L^- 1 ). The morphologic photos of all groups were observed and analysis were made. The contents of cardiomyocytes ROS and ET-lwere tested after 30 min of hypoxia and 40 min of reoxygenation, RESULTS Being injured by hypoxia and reoxygenation, refracting power of the cardiomyoeytes declined,cardiomyoeytes beat became weak or stopped. The contents of ROS and ET-1 were both obviously increased in H/R group ( P〈0. 05, compared to the control group). While compared with H/R group, the contents of ROS and ET-1 were sibnificantly decreased in BEM groups(0. 038 g ·L^- 1,0. 11 g ·L^- 1, 0. 34 g ·L^- 1 ). CONCLUSION BEM has obviously protective effect in cultured neonatal rat cardiomocytes injured by hypoxia/reoxygenation, and its mechanism may be related to the decrease of contents of ROS,ET-1.
