摘 要:目的:建立测定大鼠血浆中氯化两面针碱的高效液相色谱法,并研究氯化两面针碱与大鼠血浆蛋白的结合率。方法:平衡透析法结合高效液相色谱法,对氯化两面针碱的大鼠血浆蛋白结合率进行测定。结果:氯化两面针碱与内标完全分离,血浆中其他成分无干扰,在0.0319~2.0400mg·L-1范围内线性关系良好,低、中、高3种不同质量浓度的方法回收率>95%,日内、日间精密度均符合方法学要求。低、中、高3种药物浓度的血浆蛋白结合率分别为68.20%,68.81%,67.37%。结论:高效液相色谱法用于氯化两面针碱的生物样品测定简便、快速、准确、可靠。氯化两面针碱在大鼠血浆中具有中等强度的蛋白结合率,蛋白结合率与透析液的药物浓度无关。OBJECTIVE An HPLC method was developed to determine the concentration of nitidine chloride in rat plasma and to study the rat plasma protein binding rate of nitidine chloride. METHODS The equilibrium dialysis combined with HPLC to determine the plasma concentration of nitidine chloride and plasma protein binding rate of nitidine chloride was carried out in rat plasma. RESULTS Nitidine chloride and the internal standard were separated completely. The calibration curves of nitidine chloride was linear in the range of 0. 031 9 - 2. 040 0 mg · L^-1. Its recoveries were more than 95%, intra-day and inter-day precision of low-, mid, high-concentrations of nitidine chloride in plasma met the requirements of methodology. The rat plasma protein binding rates of nitidine chloride at low,middle and high concentrations were 68. 20%,68. 81% ,67. 37% respectively. CONCLUSION The method is sensitive, accurate and reproducible. The protein binding rate of nitidine chloride with rat plasma is of medium strength. There was no significant difference among low,middle and high concentrations.
