针刺对乳腺增生大鼠乳腺组织P53蛋白表达的影响

摘　　要：目的通过观测乳腺增生大鼠及针刺治疗后乳腺组织中P53蛋白表达的干预作用,探讨针刺对乳腺增生病的疗效与作用机理,为临床针刺治疗乳腺增生病,预防乳腺癌发生提供实验依据。方法将36只健康雌性大白鼠随机分为3组,空白对照组、模型组、针刺治疗组,每组12只。除空白对照组外,均采用贾氏造模方法建立乳腺增生模型。空白对照组、模型组大鼠于第31天开始每日抓拿一次,不作任何治疗;针刺组大鼠在第31天开始行穴位针刺治疗。治疗30天后处死大鼠,观测乳房形态大小,并采集乳腺组织标本分别固定后,采用免疫组化法测定乳腺组织的P53表达水平。结果实验结果显示模型组造模成功,细胞结构中P53表达明显;而针刺治疗组乳腺组织中P53明显轻于模型组,两者比较有显著性差异。结论针刺对乳腺增生大鼠乳腺组织P53表达有干预作用,说明针刺对乳腺增生病有治疗作用,对乳腺癌发生有预防作用。Objective By observing the rats with mammary glands hyperplasia and the intervention effects of acupuncture on the expression of P53 protein in mammary tissues, we tried to probe into the efficacy and mechanism of acupuncture on hyperplasia of mammary glands, and to provide experimental basis for acupuncture in treating mammary glands hyperplasia, and preventing the occurrence of mammary cancer. Methods 36 healthy female rats were randomized into 3 groups： the blank control group, the model group, and the acupuncture group, 12 rats in each group. Except the blank control group, the other two groups were made mammary glands hyperplasia model with Jia Shi modeling method. The rats in the blank control group and model group were grabbed once a day from the 31st day without any treatment; the rats in acupuncture group were treated with acupuncture from the 31st day. The rats were killed after 30 days of treatment. We observed the size of mammary glands, collected the samples of mammary glands and fixed. The expression of P53 in mammary tissues was determined with immunohistochemical method. Results The results of the experi- ment showed that the model was made successfully, and the expression of P53 was obvious; the expression of P53 in acupuncture group was less obvious than that of the model group, and the difference was significant. Conclusion Acupuncture has an intervention effect on the expression of P53 in mammary tissues of rats with mammary glands hyperplasia, which indicates that acupuncture has therapeutic effect on mammary glands hyperplasia, and has preventive effect on the occurrence of mammary cancer.
