摘  要:[目的]为深入研究坛紫菜的光合生理奠定基础。[方法]以坛紫菜丝状体为材料,用蔗糖密度梯度超速离心法分离纯化类囊体膜和光系统Ⅱ(PSⅡ)颗粒。[结果]色素带B1的吸收峰分别位于419、498、540、565和676 nm,色素带B2、B3、B2-a、B2-b、B3-a、B3-b和B3-c的吸收峰分别位于419、436、485和676 nm。类囊体膜B2的放氧速率和DCIP光还原活性分别为12μmol(O2)/[mg(Chl).h]和2.3ueq/[mg(Chl).h];B3的分别为37μmol(O2)/[mg(Chl).h]和25 ueq/[mg(Chl.h)];B3-b的分别为567μmol(O2)/[mg(Chl).h]和168ueq/[mg(Chl).h];B3-c的分别为8.1μmol(O2)/[mg(Chl).h]和23 ueq/[mg(Chl).h];B2-b只有DCIP光还原活性(168 ueq/[mg(Chl).h])。[结论]该研究为坛紫菜丝状体类囊体膜和光系统Ⅱ的分离鉴定提供了试验依据。[ Objective ] The purpose was to lay a foundation for researching the photosynthetic physiology of Porphyra haitanesis thoroughly. [ Method] With the conchocelis of P. haitanesis as material, the thylakoid membranes and photosystem Ⅱ (PS Ⅱ ) particles were isolated and purified by sucrose density gradient uhracentrifugation. [ Result] The absorption peaks of pigment band B1 were at 419,498,540, 565 and 676 nm resp. and that of B2, B3, B2-a, B2-b, B3-a, B3-b and B3-c were at 419,436,485 and 676 nm resp. The oxygen release rate and DCIP photoreduction activity of thylakoid membrane B2 were 12 μmol (02)/[ mg(Chl)·h] and 2.3 ueq/[ rag(Chl) ·h] resp., that of B3 were 37 μnol (O2)/[mg(Chl) ·h] and 25 ueq/[mg(Chl)·h] resp., that of B3-b were 567 tunol (O2)/[mg(Chl)·h] and 168 ueq/[mg(Chl)·h] resp., that of B3-c were 8.1 μmol ( O5 )/[ mg(Chl) .h] and 23 ueq/[ mg(Chl) -hi resp. and B2-b only had DCIP photoreduction activity ( 168 ueq/[ mg(Chl) ·h] ). [ Conclusion] The research provided test basis for isolating and identifying the thylakoid membranes and PSII from the con- chocelis of P. haitanesis.
