摘 要:目的研究补虚化瘀祛痰饮组方对尘肺病人的抗氧化作用机制。方法根据GBZ 70—2002《尘肺病诊断标准》,选取尘肺病患者120例,按随机抽样的原则分为中药干预组60例,对照组60例。测定治疗前后超氧化物歧化酶(SOD)活力、血清脂质过氧化物(LPO)含量以及周围血单个核细胞MnSOD基因mRNA的表达水平。结果对照组患者治疗前后血浆SOD活力及LPO含量差异无显著性(P>0.05),而中药干预组患者治疗后血浆SOD活力显著升高,LPO含量显著降低(P<0.01);中药干预组MnSOD基因mRNA表达水平显著高于对照组(P<0.01)。结论补虚化瘀祛痰饮可能诱导单个核细胞MnSOD基因表达,提高SOD活力,降低LPO含量,维持尘肺病人的氧化和抗氧化平衡,阻断脂质过氧化的连锁反应,干预氧自由基的产生并增强消除氧自由基的功能,可能是该方剂治疗尘肺病患者有效的机制之一。Objective To investigate the antioxidant mechanism of Buxuhuayuqutanyin for pneumoeoniosis. Methods 120 patients with pneumoeoniosis diagnosed by the diagnostic standard were randomly assigned to the treatment group (60cases) and the control group (60 cases). With the biochemical and the molecular biological techniques, superoxide dismutase (SOD)activity, lipid peroxide(LPO) level and mRNA expression level of manganese-containing superoxide dismutase (MnSOD) gene in peripheral blood mononuclear cells of patients in two groups pre and post treatment were determined. Results In the control group, there was no significant difference on serum levels of SOD and lipid peroxide (LPO) between pre and post treatment ( P 〉 0. 05 ). While in the treatment group, the activities of SOD increased significantly and the levels of LPO decreased obviously (P 〈 0. 01 ) post treatment. The results also indicated mRNA expression of mononuclear cell MnSOD gene in the treatment group was significandy higher than that of the control (P 〈0.01 ). Conclusion The results show that Buxuhuayuqutanyin can induce the expression of MnSOD gene of mononuclear cell, enhance the activity of SOD but decrease the level of LPO. It indicates that Buxuhuayuqutanyin can maintain the balance of oxidant/antioxidant, block the chain reaction of lipid peroxidation and interfere in the production of free radical and enhance the function of eliminating oxygen free radical. Antioxldation may be one of the mechanisms for this prescription to effectively treat pneumoconiosis.
