摘 要:为高效地制备高免疫活性的抗体片段,我们试用了两种新的蛋白酶消化鼠源性单克隆抗体(mAb)。纯化的IgG1抗肺癌mAb经无花果蛋白酶和菠萝蛋白酶消化后,抗体片段的产率分别为最大理论产率的77.6%和98.8%。两种酶制备的片段为F(ab′)2与Fab混合物。经细胞结合实验、细胞结合抑制实验及动物模型的肿瘤定位显像证明,两种酶所得片段均具有良好的免疫活性,其中无花果蛋白酶所制片段的免疫活性较高。To prepare the fragments of mAb with high immunoactivity efficiently, two new enzymes which can digest IgG into fragments were selected in this experiment. Ficin and bromelain could digest mAb into a mixture of F(ab′)2 and Fab within 4 h. The yields of fragments were 77.6% and 98.8% of the theoretical maximum yield respectively. The immunoreactivity of two protasegenerated fragments was tested using the solidphase radioimmunoassay. The results showed that the fragments generated by ficin and bromelain retained immunoreactivity of the mAb well. So the new enzymatic methods are of high efficiency and are convenient to perform.
