HPLC法同时测定天麻头风灵胶囊中原儿茶酸紫丁香苷和绿原酸的含量

摘　　要：目的建立同时测定天麻头风灵胶囊中原儿茶酸、紫丁香苷、绿原酸含量的方法。方法采用HPLC法。色谱柱为Diamonsil C18柱（200mm×4．6mm，5μm），流动相为甲醇-乙腈-0．05％磷酸溶液（体积比20：5：174），流速为1．0mL·min^-1，柱温为35℃，检测波长为281nm。结果原儿茶酸、紫丁香苷、绿原酸质量浓度分别在2．40—24．0mg·L^-1、5．20～52．0mg·L^-1、26．6-266mg·L^-1内与峰面积呈良好的线性关系，r分别为0．9996、0．9997、0．9995，平均回收率分别为101．0％（RSD=0．86％，n=9）、100．3％（RSD=1．9％，n=9）、101．5％（RSD=1．4％，n=9）。结论HPLC法可用于天麻头风灵胶囊的质量控制。Objective To develop a HPLC method for the determination of protocatechuic acid, syringin and chlorogenic acid in Tianma toufengling （traditional Chinese medicines） capsules. Methods The chromatographic separation was achieved on a Diamonsil C18 （200 mm × 4. 6 mm,5 μm）column with a mobile phase of methanol-acetonitrile-0. 05% phosphoric acid solution （ V： V： V = 20 ： 5 ： 174 ）. The flow rate was 1.0 mL· min^-1 and the column temperature was 35℃. Results The calibration curve was linear within the range of 2. 40 - 24.0 mg· L^-1 （ r = 0. 999 6） for protocatechuic acid,5.20 - 52. 0 mg· L^-1 （ r = 0. 999 7 ） for syringin and 26. 6 - 266 mg· L^ -1 （ r = 0. 999 5） for chlorogenic acid, respectively. The recoveries of protocatechuic acid, syringin and chlorogenic acid were 101.0% （RSD = 0. 86 %, n = 9 ）, 100. 3 % （RSD = 1.9%, n = 9）and 101.5 % （RSD = 1.4 %, n = 9 ）, respectively. Conclusions The method is accurate, sensitive and specific. It can be used for the quality control of Tianma oufengling capsules.
