大肠黏膜腺管开口中PCNA mRNA和survivin mRNA表达及与增殖、凋亡相互关系的研究

摘　　要：目的探讨增殖细胞核抗原(PCNA)mRNA和survivin mRNA在大肠黏膜各型腺管开口中的表达,以及增殖、凋亡在大肠黏膜腺管开口变化过程中动态变化关系及意义。方法采用逆转录聚合酶链反应(RT-PCR)方法提取70例不同腺管开口类型组织PCNA mRNA和survivin mRNA,并通过流式细胞学检测其增殖及凋亡指数。结果PCNA mRNA和survivin mRNA分别在Ⅰ、Ⅱ、ⅢL、Ⅳ、Ⅴ型中阳性表达率分别为0%、12.5%、38.9%、66.7%、92.3%,0%、12.5%、22.2%、44.4%、69.2%,增殖指数与凋亡指数在Ⅰ、Ⅱ、ⅢL、Ⅳ、Ⅴ型中分别为0.27±0.038、0.39±0.054、0.44±0.044、0.62±0.059、0.63±0.168,7.53±2.24、9.11±3.33、11.66±0.72、14.11±1.15、10.26±2.15。结论PCNA mRNA和survivin mRNA阳性表达在腺管开口变化过程中均逐渐增高,在Ⅰ、Ⅱ、ⅢL、Ⅳ阶段中增殖、凋亡指数同时上升,而在Ⅴ型开口中,增殖指数继续上升,而凋亡指数下降。增殖、凋亡动态平衡紊乱导致腺管开口的变化,在大肠癌发生、发展中起重要作用。Objective To study the expression of PCNA and survivin, and the relationship and significance of hyperplasy and apoptosis in the pit patterns of colorectal neoplastic. Method Expression of survivin mRNA and PCNA was detected by RT - PCR and PCR in 70 cases. Proliferation and apoptotic index were detected by flow cytometry. Result The expressions of PCNA and survivin mRNA in type Ⅰ、Ⅱ、ⅢL、Ⅳand Ⅴ were 0%, 12.5% , 38.9%, 66.7%, 92.3%, 0% .12.5% ,22.2% ,44.4% ,69.2% ,and proliferation,apoptotic index detected in type Ⅰ、Ⅱ、ⅢL、Ⅳand Ⅴ were 0.27 ± 0. 038,0.39± 0. 054,0.44 ± 0.044,0.62 ± 0. 059,0.63 ± 0. 168 ,7.53 ±2.24, 9.11 ± 3.33,11.66 ± 0.72,14. 11 ± 1.15,10.26± 2.15. Conclusions The expressions of PCNA and surviving mRNA increased gradually with both pit patterns of colorectal neoplastic. The proliferation and apoptotie index were both increased gradually in type Ⅰ、Ⅱ、ⅢL、Ⅳ, but the apoptotie index were down in type V. The presence of the out of balance of the cell proliferation and the apoptosis was confirmed significance in the course of the large intestine carcinogenesis.
