摘 要:目的观察急性百草枯(PQ)中毒大鼠肺组织中血栓调节蛋白(TM)和内皮细胞蛋白C受体(EPCR)基因表达的变化,以探讨丁M和EPCR在PQ致肺损伤中的作用。方法将40只雄性SD大鼠随机分为正常对照组(NC组,8只)和PQ染毒组(PQ组,32只,腹腔注射1%的PQ溶液20mg/kg),分别检测肺组织中TM和EPCR mRNA的表达水平,并观察PQ组大鼠的中毒表现。结果PQ组大鼠在染毒后6h和第一天时肺组织TM和EPCR mRNA的表达水平均较NC组增强(均P〈0.05或0.01);PQ组大鼠在染毒后第一天时上述指标水平已开始下降,至第七天时降至正常。结论急性PQ中毒大鼠肺组织TM和EPCR mRNA的表达水平明显上调,肺组织TM和EPCR mRNA的表达增强可能是PQ引起急性肺组织损伤的机制之一。Objective To investigate the expression of thrombomodulin(TM)and endothelial protein C receptor (EPCR) in lung tissue of acute paraquat-poisoned rats. Methods Forty male SD rats were randomly divided into two groups:normal control group (NC, n=8) and PQ group (n=32, 20mg/kg 1%PQ i.p). The expression of TM mRNA and EPCR mRNA in lung tissue were detected by RT-PCR. The ethology of rats was observed after PQ poisoned. Results Compared with that in normal control group, the expression of TM mRNA and EPCR mRNA in lung tissue of PQ group markedly increased at 6h (P〈0.0.5 or 0.01), decreased slowly at the 1st day and returned to normal levels at the 7th day. Conclusion The expression of TM and EPCR genes increases in rats after paraqual poisoning, which indicates that TM and EPCR genes may be involved in lung injury.
