同种异体移植的骨髓间充质干细胞在椎间盘环境中存活及增殖能力的研究

摘  要:目的观察同种异体移植的骨髓间充质干细胞(MSCs)在椎间盘环境中的存活及增殖能力。方法从30只日本大白兔中分离培养骨髓MSCs,Ad—EGFP标记后同种异体移植到椎间盘环境中,术后1周、2周、4周、8周、12周取材,Brdu处理后做石蜡切片,荧光显微镜观察细胞存活,并行Brdu免疫组化染色检测细胞增殖。结果术后1周、2周、4周、8周、12周标本内可观察到绿色荧光,术后1周、2周、4周、8周、12周标本内均有Brdu免疫组化染色阳陆细胞。结论自体移植的MSCs在椎间盘环境中能够存活并且增殖,为MSCs作为椎间盘组织工程的种子细胞提供了实验依据。Objective To observe the viability and proliferation of allografted messenchymal stem cells in the intervertebral disc. Methods Bone mesenchymal stem cells isolated from 12 Japanese white rabbits were labeled with Ad-EGFP and were allografted into lumbar intervertebral disc of which the nucleus pulposus had been aspirated. The lumbar spine were extracted 1, 2, 4, 8, 12 weeks after the operation, dealed with Brdu, the viability and proliferation of the cells were assessed with consecutive sections using fluorescent microscope and immunohistochcmistry of Brdu. Results Fluorescence was observed in the specimens for 1, 2, 4, 8, 12 weeks after the operation and positive immunohistochemical staining for Brdu was observed in the specimens for l, 2, 4, 8, 12 weeks after the operation. Conclusion Allografted mesenchymal stem cells can survive and proliferate in the intervertebral disc. Results from our study provide experimental evidence for MSCs as seeding cells of intervertebral disc tissue engineering.
