高糖对人近端肾小管上皮细胞TGF-β1、PAI-1基因表达的影响

摘  要:目的观察高糖(HG)对人近端肾小管上皮细胞(HKC)分泌转化生长因子β1(TGF-β1)和纤溶酶原激活物抑制因子1(PAI-1)mRNA表达水平的影响。方法采用数字表法随机分为低糖组(5.5mmol/L,LG组)和HG组(25mmol/L),分别培养于HKC中,于培养0、24和48h时收获细胞,并采用RT-PCR法检测两组HKC分泌TGF-β1和PAI-1 mRNA的表达水平。结果HG组培养24h和48h时的HKC分泌TGF-β1和PAI-1 mRNA表达水平均较LG组明显升高(均P〈0.01)。且两组培养48h时的上述指标均较24h时明显升高(均P〈0.O01);经Spearman相关分析显示,HKC分泌PAI-1和TGF-β1 mRNA的表达水平之间存在正相关性(r=0.802,P〈0.01)。结论HG可刺激HKC分泌TGF-β1和PAI-1 mRNA表达水平的增高。Objective To investigate the effect of high glucose concentration on synthesis of TGF-β1 and PAI-1 mRNA in cultured human proximal tubular epithelial cells. Methods Human proximal tubular epithelial cells cultured with high glucose (25mmol/L, HG)or low glucose (5mmol/L, LG)concentrations,the expression of TGF-β1 and PAI-1 was assessed by RT-PCR. Results TGF-β1 and PAI-1 mRNA levels were significantly increased in HG groups compared with LG groups. Conclusion High glucose can significantly up-regulate TGF-β1 and PAI-1 gene expression in cultured human proximal tubular epithelial cells.
