健脑益智胶囊对脑出血大鼠血清及脑组织中SOD和MDA的影响

摘　　要：目的探讨健脑益智胶囊对脑出血大鼠脑保护作用的机制。方法采用自体血注入法建立大鼠脑出血模型。将50只SD大鼠按照随机原则分为假手术组、模型组及健脑益智胶囊小、中、大剂量治疗组（分别为0．5、1．0、2．0g／kg，每日1次），均给药14d。用Longa评分法和Bederson评分法观察各组大鼠行为学评分变化。术后14d处死大鼠，分别检测血清和脑组织中超氧化物歧化酶（SOD）和丙二醛（MDA）水平。结果药物治疗14d，大鼠运动能力的改善与治疗药物的剂量呈正相关，大剂量组Bederson评分显著低于假手术组和模型组（P均〈0．05）。药物治疗大剂量组血清和脑组织中SOD活性均高于其他各组，MDA含量均低于其他各组，与假手术组和模型组比较差异均有统计学意义（P均〈0．05），与小、中剂量治疗组比较差异无统计学意义（P均〉0．05）。结论健脑益智胶囊能升高脑出血模型大鼠血清和脑组织中SOD活性，降低MDA含量，对脑出血后氧自由基的清除有明显作用。Objective To approach the protective mechanism of Jiannao Yizhi capsule （健脑益智胶囊） on the brain in rats with cerebral hemorrhage. Methods The cerebral hemorrhage model was induced by injection of auto-blood. Fifty Sprague-Dawley （SD） rats were randomly divided into sham operation group, model group and three treatment groups.. Jiannao Yizhi capsule low-, middle- and high-dose groups （0.5, 1.0 and 2.0 g/kg, once a day）. The therapeutic course was 14 days in each group. Longa and Bederson＇s scores were used to evaluate the score of rats＇ behavior or ethology in each group. The rats were executed 14 days after the operation. The serum and brain tissue levels of superoxide dismutase （SOD） and malondialdehyde （MDA） were measured respectively. Results The level of SOD in high-dose group was significantly higher and the level of MDA and Bederson＇s score at 14 days, obviously lower than that respectively in the other groups, compared with the sham operation group and model group, the differences being statistically significant （P〈0.05）, and compared with the middle- and low-dose groups, the differences being of no statistical significance （all P〉0. 05）. Conclusion Jiannao Yizhi capsule can elevate SOD and decrease MDA levels in the serum and brain tissue of rat model with cerebral hemorrhage, and possesses marked effect on clearing the oxygen free radical after cerebral hemorrhage.
