摘 要:目的研究黄蜀葵总黄酮(TFA)对脑卒中后抑郁(PSD)大鼠下丘脑CRF表达的影响。方法线栓法制备局灶性脑缺血大鼠模型,复合孤养和应激制备PSD大鼠模型。观察PSD大鼠糖水消耗量的变化,应用免疫组化法检测PSD大鼠下丘脑CRF阳性神经元的平均光密度,应用逆转录聚合酶链反应检测下丘脑CRFmRNA的表达。结果TFA(50、100 mg.kg-1,ig,qd×24d)可明显增加PSD大鼠糖水消耗量,降低PSD大鼠CRF阳性神经元的平均光密度值,并抑制下丘脑CRFmRNA的过度表达。结论TFA可显著降低PSD大鼠下丘脑CRF和CRFmRNA的过度表达,其机制可能与TFA抑制HPA轴的过度亢进有关。Ahn To study the effects of total flavone of A(TFA) on the expression of corticotropin releasing factor(CRF) and mRNA in the hypothalamus of rats with post stroke depression(PSD). Methods Middle cerebral artery occlusion (MCAO)followed by isolation feed and irritable stimuli was used to induce PSD model in rats. The changes of sucrose drinking of PSD rat model were investigated. CRF and CRFmRNA were assayed by immunohistochemistry and RT-PCR. Results TFA (50,100 mg ·kg-1,ig,qdx24d) could signifi- cantly increase the consumption of sucrose solution, decrease the mean optical density values of CRF positive neurons, downregulate the excessive expression of CRFmRNA. Conclusion TFA can obviously decrease the over-expression of CRF and CRFmRNA in rats with PSD, the mechanisms of which might be associated with its inhibition effect on the activating of HPA axis.
