摘  要:目的观察蕨麻正丁醇提取部位(NP)对垂体后叶素致小鼠急性心肌缺血后天冬氨酸特异性半胱氨酸蛋白酶9/3(caspase-9/3)mRNA表达的影响。方法90只雌性昆明种小鼠被随机分为正常对照组、缺血模型组、复方丹参组和NP高、中、低剂量组,每组15只。各组连续灌胃NP浸膏羧甲基纤维素钠溶解物25d,于末次给药1h后用腹腔注射垂体后叶素(20U/kg)建立小鼠急性心肌缺血模型。监测制模前后心电图Ⅱ导联J点变化;测定血清乳酸脱氢酶(LDH)活性;用逆转录-聚合酶链反应(RT—PCR)检测心肌组织caspase-9/3的mRNA表达。结果复方丹参组和NP高、中、低剂量组缺血所致心电图J点的上移均较缺血模型组显著降低[(0.100±0.055)、(0.105±0.038)、(0.111±0.034)、(0.145±0.016)mV比(0.223±0.043)mV,P均〈0.01],复方丹参组及NP高、中剂量组可显著降低缺血损伤小鼠血清LDH活性[(4277.160±878.327)、(2650.129±917.367)、(4752.069±765.974)U/L比(5769.319±826.513)U/L,P均〈0.01]。复方丹参组和NP高、中、低剂量组小鼠心肌caspase-9mRNA表达均较缺血模型组显著下调(0.461±0.103、0.227±0.056、0.435±0.089、0.876±0.094比1.341±0.102,P均〈0.01);正常对照组、复方丹参组及NP高剂量组小鼠心肌caspase-3mRNA表达呈阴性,而缺血模型组及NP中、低剂量组表达明显。结论NP具有对抗急性心肌缺血损伤的作用,其机制可能与抑制凋亡相关基因caspase-9/3的表达有关。Objective To study the effects of n-butanol extract of Potentilla anserina L. (NP,蕨麻) on the expressions of caspase-9/3 mRNA in mice with pituitrin-induced injury of acute myocardial ischemia. Methods Ninety healthy mature female mice were randomly divided into normal control group, untreated (ischemic) group, Salvia rniltiorrhiza (复方丹参) group and high-dose, medium-dose and low-dose NP groups (each group, n= 15). Mice were treated by intragastric administration with NP extractum and sodium carboxymethycellulose (CMC-Na) for 25 days. Acute myocardial ischemic model was induced by intra-peritoneal administration of pituitrin (20 U/kg) in mice 1 hour after last intragastric administration. Before and after the model was established, electrocardiogram in mice was monitored and the height of the J spot was measured. The activities of lactate dehydrogenase (LDH) in serum of mice were detected. The expressions of caspase-9/3 mRNA were assayed by reverse transcription-polymerase chain reaction (RT-PCR). Results Electrocardiogram: the heights of J spots shifted in Salvia miltiorrhiza group, high-dose, medium-dose and low-dose NP group were all significantly lower than the height in untreated group ((0. 100±0. 055), (0, 105±0. 038), (0. 111±0. 034) and (0. 145±0. 016) mV vs. (0. 223±0. 043) mV, all P〈0. 01]. Serum biochemical indicators: compared with untreated group, the activities of LDH was decreased significantly in Salvia miltiorrhiza and high- and medium-dose NP groups [(4 277. 160±878. 327), (2 650. 129±917. 367) and (4 752. 069 ± 765. 974) U/L vs. (5 769. 319± 826. 513) U/L, all P〈0. 01]. RT-PCR: the expressions of caspase-9 mRNA in cardiac muscles of Salvia miltiorrhiza group, high-dose, medium-dose and low-dose NP groups were all significantly down-regulated compared with the expression in the untreated group (0. 461±0. 103, 0. 227±0. 056, 0. 435±0. 089, 0. 876±0. 094 vs. 1. 341±0. 102, all P〈 0. 01).
