摘  要:目的探讨米诺环素对脑缺血-再灌注大鼠认知功能障碍及能量代谢的影响。方法利用线栓法造成大脑中动脉栓塞的局灶性脑缺血,制备大鼠学习记忆功能障碍模型。采用跳台实验和Y型迷宫实验测试大鼠学习记忆能力,采用反相高效液相色谱法检测脑组织三磷腺苷(ATP)含量、腺苷酸池(TAN)水平和能量负荷值(EC);试剂盒检测乙酰胆碱(Ach)含量。结果米诺环素明显改善脑缺血-再灌注所致认知功能障碍;提高脑组织中ATP含量、TAN水平和EC值及Ach含量。结论米诺环素能改善脑缺血-再灌注大鼠认知功能障碍,其作用机制可能与改善脑能量代谢有关。Objective To investigate the effects of minocycline on study and memory dysfunction and energy metabolism after temporary middle cerebral artery occlusion (tMCAO) in rats. Methods The model of study and memory dysfunction was established after 90 min of MCAO in rats. The study and memory capability was tested by jumping stand the Y- shape maze experiments. Total adenine nucleotides (TAN), energy charge (EC) and ATP levels were measured by RP-HPLC. Contents of acetylcholine (Ach) was detected by the kit. Results The severe neuronlogic deficits, study and memory dysfunction induced by tMCAO were significantly improved after the treatment of minocycline; meanwhile, levels of TAN, EC, ATP and Aeh were significantly enhanced. Conclusion Minocycline can improve the study and memory function by ameliorating the energy metabolism.
