摘 要:目的观察金银花主要有效成分绿原酸体外抗人巨细胞病毒(HCMV)的效果和安全性。方法用细胞病变法和噻唑蓝(MTT)法检测绿原酸抗HCMV的最大无毒浓度、最小有效浓度和治疗指数,并与更昔洛韦、金叶败毒及其君药金银花进行比较。结果更昔洛韦、金叶败毒、金银花及绿原酸的最大无毒浓度分别为10,3 000,3 000及100μg.mL-1,最小有效浓度分别为10,300,3 000及1μg.mL-1,治疗指数分别为1,10,1及100。结论绿原酸细胞毒性略高于金叶败毒和金银花,但抗HCMV效果明显高于金叶败毒和金银花,治疗指数最高,具有抗HCMV的潜在优越性。Objective To study the in vitro anti-cytomegalovirus effect and the security of chlorogenic acid. Methods The 0% toxic dose (TD0 ), minimum effective concentration (MEC) and therapeutic index (TI) of anti- cytomegalovirus activity by chlorogenic acid were detected with cytopathic assay and MTT method. Ganciclovir, Jinyebaidu (JYBD) and honeysuckle were used as the control drugs for comparison. Results The TD0 of ganciclovir, JYBD, honeysuckle and chlorogenic acid were 10, 3 000, 3 000 and 100 μg·mL-1 , the MEC were 10, 300, 3 000 and 1 μg·mL-1 , TI were 1, 10, 1 and 100, respectively. Conclusion The cytotoxicity of chlorogenic acid is more obvious than that of JYBD and honeysuckle, but the anti-cytomegalovirus effect is more effective, and it has the highest TI. Therefore, chlorogenic acid has the potential advantages of anti-cytomegalovirus effect.
