摘  要:用EMS(甲基磺酸乙酯)诱变处理海沃德与红阳猕猴桃叶片,进行定向筛选耐盐突变体研究。结果表明,在相同EMS浓度处理下,胚性愈伤组织随着处理时间的增加发生的比率降低;在同一处理时间下,胚性愈伤组织随着EMS浓度增加发生的比率降低。当诱变率达50%致死剂量时,EMS处理浓度为6%、处理时间为2 h时,对海沃德与红阳猕猴桃的处理效果最佳,其后代的分化率可达到70%以上;在含NaCl 1.0 g/L的培养基上通过胚性愈伤组织诱导出海沃德和红阳的组培苗,海沃德11株,红阳组培苗10株,经继代培养筛选鉴定,诱变处理所得植株与正常培养植株相比,其耐盐性明显提高。Research on leaves with EMS (eth filter oriented salt-tolerant mutant from the Redsun and Hayward kiwifruit yl methane sulphonate) mutagenesis treatment in this paper. The results showed that the formation rate of embryogenic calli redued by increase processing time at the same EMS concentrations and at the same time treatment. 6 % concentration of EMS and 2 h treatment with Redsun and Hayward kiwifruit leaves got the best result at semi-lethal dose. Differentiation rate of the Redsun and Hayward reached more than 70% ; We got Hayward tissue plants 11 and the Redsun 10 by 1.0 g/L NaCl on the medium from the embryogenic callus. Compared with the normal cultivate plants, cultivated by directional filter plant got significantly improve their salt tolerance, through physiological and biochemical identification.
