摘  要:目的:研制调胃承气汤证兔动物模型。方法:将60只日本大耳白兔随机分为正常对照组、调胃模型组、模型治疗组和模型验证组。在次碳酸铋、禁水的基础上,用大肠杆菌内毒素攻毒,观察家兔的体温、腹围、二便等一般状态、检测白细胞数量和NO含量、脏器的病理形态学变化。结果:模型组家兔体温升高,腹围增大,WBC和NO含量明显增高(P(0.01),部分脏器出现了明显的病理改变。治疗组和验证组则有明显的改善。结论:采用这种多因素联合造模方法研制出的调胃承气汤证兔动物模型,在症状、体征、实验室指标及病理改变上与调胃承气汤证有较强的相关性,可以认为该造模方法是比较成功的。Objective: To develop animal models that could be adequately used to show the syndromes suitable for Tiaoweichen- gqi Decoction. Methods :The study was carried out with 60 lab rabbits by equally and randomly dividing the animals into 4 groups. The groups included Control, Pathological Model of Tiaoweichengqi Decoction, Model-Treating of Tiaoweichengqi Decoction, Mod- el-Proving of Tiaoweichengqi Decoction. Amounts of bismuth subcarbonate were given to the Model groups for 2 days, no water for 24 hours on the second day, then intravenously injected E. coli endotoxin. During the experiment, a rabbit's spirit, body tempera- ture and abdominal circumference were observed or measured regularly. Blood samples were collected for testing WBC and NO. Pathological changes in lung, liver, intestines and kidney were examined under microscope. Results : There were obvious changes of temperature and abdominal circumference in the Pathological Model group. There were significant differences ( P 〈 0. 01 ) on the WBC counts and the contents of NO between the Control and Model groups. Under the microscopic, some changes found in the rabbits in the Pathological Model group. Conclusion:The multifactor intervention approach with some dosages of bismuth subcarbonate ,water deprivation and endotoxin injection could create the major symptoms ,physiological signs and pathological changes in lab rabbits strikingly close to what described by the TCM theories. Thus, this method of developing models seemed appropriate for the purpose.
