2型糖尿病大鼠胰岛素受体底物-2基因表达变化及中药的干预作用

摘　　要：目的:观察2型糖尿病大鼠血糖、血清胰岛素浓度和胰岛素受体底物-2(insulin receptor substrate 2,IRS-2)基因表达的变化及复方黄连降糖片对其变化的影响。方法:以小剂量链脲佐菌素(28mg/kg)腹腔注射合并高脂饮食诱导2型糖尿病大鼠模型,给予不同剂量复方黄连降糖片治疗8周。处死后分别测定其血糖、血清胰岛素浓度,肝脏、骨骼肌和脂肪组织的IRS-2的mRNA及蛋白表达。结果:2型糖尿病大鼠的血糖、血清胰岛素升高,各组织IRS-2的mRNA及蛋白表达下降;复方黄连降糖片能降低模型大鼠的血糖和血清胰岛素,并能显著升高各组织IRS-2的mRNA及蛋白表达。结论:复方黄连降糖片治疗2型糖尿病可能与其提高外周组织IRS-2的基因表达从而改善胰岛素的信号传导有关。Objective： To investigate blood glucose, insulin, the gene expression of insulin receptor substrate - 2 （ abbr： IRS - 2） of type 2 diabetes mellitus（abbr：T2DM） rats and effects of Fufang huanglian jiangtangpian （ abbr： FFHLJTP） on these indexs. Method~：Model rats of T2DM were induced by intraperitoneal injection of low dose streptozotocin and high fat-diet. Model rats were treated with different dose FFHLJTP for eight weeks. All rats were killed and followed indexs were measured： blood glucose, insuhn, expression of IRS -2mRNA and protein in liver, muscle and fat. Results：The blood glucose, insulin of T2DM rats are in- creased, and expression of IRS -2mRNA and protein in liver, muscle and fat are decreased in T2DM rats. FFHLJTP can decrease the blood glucose,insulin and increase the expression of IRS - 2mRNA and protein in liver, muscle and fat of T2DM rats. Conclusion：The mechanism of FFHLJTP treating T2DM may be partly by raising IRS -2 gene expression in liver, muscle and fat to improve insulin resistance.
