摘 要:目的对中国六省分离的伤寒沙门菌进行脉冲场凝胶电泳(PFGE)分析,初步建立中国伤寒沙门菌的数据库。方法采用PFGE技术,对中国1979-2005年六省分离的伤寒沙门菌用XbaⅠ酶切,用BioNumerics软件对PFGE图谱进行分析,并且建立中国分子分型数据库。结果收集的417株菌株共有288种不同的PFGE带型,相似性系数在55.2%~100%之间,不同的省份和年份之间有交叉带型。结论中国伤寒沙门菌的菌株变异较大,不同的省份可能存在不同克隆系的伤寒沙门菌菌株的流行。通过建立中国伤寒沙门菌PFGE带型数据库,对中国以后伤寒沙门菌的监测具有重要的意义。Objective To analyze the molecular types of Salmonella Typhi strains isolated from six provinces in China, and establish the PulseNet databank. Methods Pulsed field gel electrophoresis (PFGE) was used to analyze Salmonella Typhi strains isolated from 6 provinces during 1979-2005. The genomic DNA in different Salmonella Typhi strains were extracted after digesting with XbaⅠ . The BioNumerics software was used to analyze the PFGE patterns. Results PFGE patterns of Salmonella Typhi strains isolated in China in different years were quite different. There were 288 PFGE patterns in total. The similarity value of the strains ranged from 55.2% to 100%. The same patterns were discovered in different province and in different year. Conclusion Salmonella Typhi strains isolated in China varied, and different clones might exist in different provinces. The establishment of PulseNet database is of great significance for the surveillance of Salmonella Ttyphi strains in China.
