摘 要:目的研究高压氧对缺血再灌注损伤大鼠脑组织脑红蛋白表达的影响,探讨高压氧的脑保护作用机制。方法30只大鼠随机分为对照(假手术)组、缺血再灌注48h组(48h I/R组)及缺血再灌注48h加高压氧治疗组(48h HBO组),每组10只。采用双侧颈总动脉夹闭法制备大鼠全脑缺血再灌注损伤模型,应用免疫组织化学法检测缺血再灌注损伤后脑组织脑红蛋白的表达并用千湿重法测定脑含水量的变化。结果与对照组相比,48h I/R组及48h HBO组脑红蛋白光密度值和脑组织含水量均明显升高(P〈0.05)。48h HBO组脑红蛋白表达高于48h I/R组,脑组织含水量低于48h I/R组(P〈0.05)。结论高压氧可提高对缺血再灌注损伤大鼠脑组织脑红蛋白的表达,也是其对大鼠脑组织保护作用的机制之一。Objective To investigate the effect of hyperbaric oxygen (HBO) on the expression of neuroglobin (NGB) following cerebral ischemia-reperfusion injury in rats, and to explore related protective mechanism of HBO. Methods Thirty Wistar rats were randomly divided into 3 groups : the sham surgery group (n = 10), the 48-hour ischemia-reperfusion group (48 h I/R, n = 10) and the group treated with hyperbaric oxygen after 48-hour ischemia-reperfusion (48 h HBO, n = 10). An experimental cerebral ischemia-reperfusion injury model was developed by clamping bilateral carotid arteries for 20 minutes. The expression of NGB in the brain tissue was monitored with immunohistochemical methods, and changes in the content of brain water were measured with wet and dry weight methods. Results Compared with that of the sham surgery group, optical density (OD) of NGB and the brain water content all increased significantly in both groups (P 〈 0.05 ). The OD value of NGB in the 48 h HBO group was higher than that of the 48 h I/R group, but brain water content in the 48h HBO group decreased when compared to that of the 48 h I/R group(P 〈 0.05 ). Conclusions HBO could enhance the expression of NGB in animals following 48 h cerebral ischemia-reperfusion injury, which might be one of the protective mechanisms of HBO.
