摘 要:目的:探讨杀伤细胞免疫球蛋白样受体(KIR)基因型与白血病发病之间的关系.方法:采用序列特异性引物聚合酶链反应(PCR-SSP)法对263例白血病患者和239例健康正常对照的KIR基因多态性进行检测,比较KIR基因型频率在白血病患者和健康正常对照之间、不同白血病类型之间的差异.结果:263名白血病患者和239名正常对照中KIR2DL4,3DL2,3DL3,3DP1的基因频率均为1.000.其中KIR2DL1,2DL3,3DL,2DP1基因频率较高,而2DL5B基因频率较低.白血病患者和正常对照中KIR2DS4基因频率分别为0.5101,0.3901,2组组间差异具有统计学意义(P<0.01),其余基因频率2组组间差异均无显著性.慢性粒细胞白血病(CML)、急性非淋巴细胞白血病(NALL)、急性淋巴细胞白血病(ALL)3组患者中,KIR2DS4基因频率分别为0.6050,0.4017,0.4569;CML组KIR2DS4频率明显高于NALL,ALL组和正常对照组(P<0.05);NALL,ALL组与正常对照组之间KIR2DS4频率差异无显著性.CML,NALL,ALL组中其余基因频率差异均无统计学意义.结论:激活型KIR2DS4可能与慢性粒细胞白血病发病有一定的关联.AIM: To investigate the association of killer cell immunoglobulin-like receptors (KIR)gene polymorphisms with leukemia. METHODS: Two hundred and sixty-three patients and two hundred thirty-nine randomly selected controls were genotyped for KIR genes by PCR-SSP. RESULTS: The genotypie frequencies of KIR2DL4, 3DL2, 3DL3 and 3DP1 were 1. 000 in all the patients and controls under study. The genotypes of KIR2DL1, 2DL3, 3DL1 and 2DP1 were most prevalent genotypes while the frequency of genotype 2DLSB was very low in all the patients and controls under study. The frequency of KIR2DS4 was much higher in patients with chronic granulocytic leukemia(CML) than that in controls( P 〈 0.01 ). CONCLUSION: The activating genotype KIR2DS4 may be associated with the susceptibility of CML.
