摘 要:目的:观察Janusin不同片段作为细胞外基质时对脊髓培养神经细胞形态的影响.方法:胚胎第14日~15日龄小鼠脊髓神经细胞接种在铺有原核表达的不同Janusin片段的培养孔内,在无血清培养液中培养26h,经ABC法染色,图像处理仪摄取图像后,用体视学方法测量NSE阳性细胞数及突起长度,计算单个细胞的突起长度并做统计学方差分析.结果:在无血清培养条件下,细胞呈不同形态:一种胞体较大、折光性差,突起具有复杂的分支,数量较多;一种胞体小、折光性强,多为单极或双极,突起分支少,数量较少;还有一种胞体扁平,自胞体向四周有较均匀的放射状突起,胞质内有2个~4个圆而亮的小泡状的结构,数量最少.结论:Janusin分子上的不同片段作为细胞外基质时,对脊髓培养神经细胞的突起生长影响不同,FN1-2组平均突起长度最短,明显抑制突起生长。Aim: To study the effects of different fragments of Janusin on morphological changes of neurons in vitro . Methods: The cell suspension were prepared from E14 15 mouse embryo spinal cord by mechanical dissection and digestion with enzyme. The cells were added to the culture dish absorbed different Janusin fragments expressed by E.coli . The cells were cultured in serum free medium for 26 h. The cells were then fixed and stained by ABC immunocytochemical method for NSE. The number of the cells and the length of the neurites were measured by stereological method using an image analysis. The result was analyzed statistically. Results: The cells grew well in the serum free medium for 26 h. Three kinds of cells could be identified: bright cells with single or two neurites; dark cells with well branched neurites; flat cells with 2 ̄4 round vesicles in the cell body. The cell number and the length of neurites could be influenced by different fragments of Janusin. It was found that FN1 2 inhibited neurite growth. Conclusion: Different motifs of Janusin used as substrate have different effects on cultured neural cells, adhesion or antiadhension, promotion or inhibition of the growth of neurites. They exert their influence through receptors on the cell membrane. This study may provide some clues to search for the different receptors which may play essential roles in neuronal development and plasticity.
