摘 要:目的:研究昆布(裙带菜)提取物岩藻黄质(fucoxanthin)对7种人肿瘤细胞的生长抑制作用。方法:台盼兰拒染法测定岩藻黄质对7种人肿瘤细胞(K562、HL-60、MCF-7、HepG2、Hela、Jurkat、U937)的细胞毒作用,流式细胞术检测细胞周期及细胞凋亡率。结果:岩藻黄质对人肿瘤细胞具有显著的细胞毒作用,其中对HL-60、MCF-7、HepG2、Hela肿瘤细胞的抑制作用表现出浓度依赖性,对K562、Jurkat和U937细胞的抑制作用尚具浓度和时间依赖性特征。其对人正常肝细胞的抑制作用浓度远远高于肿瘤细胞。Fucoxanthin作用后能使K562、Jurkat和U937细胞的细胞周期阻滞于G0/G1期,对细胞周期的进程具有抑制作用,能明显诱导K562、Jurkat和U937细胞出现早期凋亡。结论:昆布有效成分岩藻黄质诱导细胞凋亡是其抑制肿瘤细胞增殖的重要途径。Objective: We assessed multiple in vitro biological activities of a main component of Thallus japonica-fucoxanthin over several tumor cell lines.Methods: Tumor cell lines were evaluated using the following tests: trypan blue exclusion test,flow cytometry,and annexin V-FITC/propidium iodide assays.We selected chronic myelogenous leukemia cells(k562),human acute promyelocytic leukemia cells(HL60)human breast adenocarcinoma cells(MCF-7),hepatoma cells(HepG2),cervix tumor cells(Hela),human T cell leukaemia cancer line (Jurkat) human histiocytic lymphoma cells(U937) and normal hepatic cell( L-02 ). Results:h showed that fucoxanthin induced significant cell growth inhibition in an dose-dependent manner in HL-60, MCF-7, HepG2, Hela cells. The results in K562, Jurkat and U937 cells showed that the inhibition effects of fucoxanthin in a dose- and time-dependent manner. And the 50% inhibition concentration of fueoxanthin in normal cell was much higher than that in tumor cells. It was noticed that growth arrest was induced by G0/G1 phase arrest and apoptotic cell death. Conclusion: The results of this study indicate that fucoxanthin exerts multiple biological activities in vitro. Fucoxanthin appears to be a good candidate to be used as an antitumor agent.
