摘 要:背景:珊瑚羟基磷灰石人工骨用于修复骨缺损在临床上已很常见,但复合抗肿瘤药物的人工骨还没有应用于临床,关于这方面的基础研究还需进一步深入。目的:研制复合抗肿瘤药物顺铂的珊瑚羟基磷灰石人工骨,并观察其体外缓释浸提液对骨巨细胞瘤原代培养细胞增殖的影响。设计、时间及地点:对比观察实验,于2006-06/11在解放军广州军区总医院骨科研究所及南方医科大学细胞生物学实验室完成。材料:珊瑚羟基磷灰石人工骨由解放军广州军区广州总医院骨科研究所提供,顺铂为济南齐鲁制药厂生产,人骨巨细胞瘤标本由解放军广州军区广州总医院骨科提供。方法:通过真空冷冻干燥等处理将顺铂载入珊瑚羟基磷灰石人工骨,以形成复合抗肿瘤活性人工骨;将复合人工骨浸入模拟体液,收集第2,4,6,8周浸提液。采用四甲基偶氮唑蓝法检测浸提液在体外对骨巨细胞瘤的增殖抑制作用。以未复合顺铂的珊瑚羟基磷灰石人工骨浸提液对照。主要观察指标:肿瘤细胞增殖抑制率。结果:第2,4,6,8周复合顺铂的珊瑚羟基磷灰石人工骨浸提液对骨巨细胞瘤原代培养细胞的增殖抑制率分别为67.9%,48.3%,34.57%和28.4%。同时提取的珊瑚羟基磷灰石人工骨浸提液对骨巨细胞瘤原代培养细胞则无明显抑制作用。结论:该复合抗肿瘤活性人工骨具有良好的缓释功能,在8周内对骨巨细胞瘤细胞仍有较好的增殖抑制作用。BACKGROUND: Coral hydroxyapatite bone graft has been widely used for repairing bone defect in clinic; however, bone graft compound with antitumor drug still needs to be further studied. OBJECTIVE: To prepare a cisplatin-impregnated coral hydroxyapatite bone graft and to observe the effect on primary-cultured cell proliferation of giant cell tumor of bone. DESIGN, TIME AND SETTING: A contrast study was performed at Institute of Orthopaedics, Guangzhou General Hospital of Guangzhou Military Command of Chinese PLA and Department of Cell Biology, Basic Medical College, Southern Medical University from June to November 2006. MATERIALS: Coral hydroxyapatite bone graft was provided by Institute of Orthopaedics, Guangzhou General Hospital of Guangzhou Military Command of Chinese PLA; Cisplatin was provided by Qilu Pharmaceutical Factory, Jinan; samples of human giant cell tumor of bone were provided by Department of Orthopaedics, Guangzhou General Hospital of Guangzhou Military Command of Chinese PLA. METHODS: The cisplatin was impregnated with coral hydroxyapatite bone graft following vacuum freeze-drying to prepare bone graft with anti-tumor activity. Thereafter, the compound bone graft was immersed in simulating body fluid to obtain leaching liquor at the 2, 4, 6 and 8 weeks. MTT method was used to detect the inhibitor effect of teaching tiuquid on proliferation of giant cell tumor of bone. Cisplatin-impregnated coral hydroxyapatite bone graft was considered as the controls. MAIN OUTCOME MEASURES: Proliferation-inhibition rate of tumor cells. RESULTS: The proliferation-inhibition rates of cisplatin-impregnated coral hydroxyapatite bone graft leaching liquor were 67.9%, 48.3%, 34.57%, and 28.4% respectively at 2, 4, 6 and 8 weeks. In addition, coral hydroxyapatite bone graft leaching liquor had no inhibitor effect on giant cell tumor of bone.
