摘 要:目的探讨低强度635nm激光照射对体外培养的大鼠骨髓间充质干细胞(bone marrow derived mesenchymal stem cells,BM-SCs)增殖及分泌功能的影响。方法体外培养的第2代SD大鼠BMSCs以InGaAsP半导体激光(635nm,60mW)不同能量密度(0,0.5,1.0,2.0和5.0J/cm2)照射,24h后酶联免疫吸附法(ELISA)测定上清液中血管内皮生长因子(VEGF)及神经生长因子(NGF)含量;BrdU法绘制BMSCs增殖曲线。结果低强度635nm半导体激光照射能够促进体外培养的大鼠BMSCs的增殖,最佳能量密度为0.5J/cm2;大鼠BMSCs能够在体外培养条件下分泌VEGF、NGF;5.0J/cm2低强度半导体激光照射能够显著促进BMSCs分泌VEGF、NGF。结论635nm波长低强度半导体激光照射能够显著促进体外培养的大鼠BMSCs增殖及分泌功能,且与照射剂量有关。Objective To investigate the influence of low-level laser irradiation (LLLI) at different energy densities on proliferation and secretion of rat bone marrow derived mesenchymal stem cells (BMSCs). Methods BMSCs used to irradiate were from second generation SD rats (passage 2, P2). A 635 nm indium-gallium-arsenate-phosphate (InGaAsP) diode laser with 60 mW optical power was used as the source of irradiation at continuous and non-contact mode. After 24 hour incubation period, production of vascular endothelial growth factor (VEGF) and nerve growth factor (NGF) in the culture medium was measured with enzyme-linked immunosorbent assay (ELISA). Cell proliferation was assessed by using 5-hromo-2'-deoxyuridine (BrdU) cell proliferation assay. Results LLLI at 635 nm wave length could affect proliferation of rat BMSCs in vitro and the energy density of 0.5 J/cm2 was found to an optimal value to significantly stimulate proliferation of BMSCs as compared to non-irradiated group and the other irradiated groups. VEGF and NGF could be secreted by BMSCs and significant stimulation of VEGF and NGF secretion was obtained with the samples irradiated at the energy density of 5.0 J/cm2. Conclusions Our results indicated that LLLI at 635 nm wave length could significantly increase proliferation and secretion of rat BMSCs in vitro in a dose-dependent manner.
