摘 要:目的探讨骨髓间充质干细胞移植于大鼠玻璃体腔和视网膜下间隙的分化情况。方法体外培养大鼠骨髓间充质干细胞并传至第五代,应用HE染色和免疫组织化学方法观察BMSCs移植于大鼠玻璃体腔和视网膜下间隙30天的生长和分化特点。结果移植后30天,玻璃体腔和视网膜下间隙新生出一层细胞,排列成条索状,形似视网膜细胞,部分细胞显示NSE、GFAP、NF200阳性。结论移植于大鼠眼内的BMSCs在眼内微环境的作用下具有向视网膜样结构分化的潜能。Objective To explore the differentiation of the BMSCs (bone-marrow mesenehymal stem cells), after the BMSCs were transplanted into the vitreous and subretinal space in rat eyes. Methods After the BMSCs were separated and purified, they were generated to the fifth generation. Microscopical observation, immtmohistochemical staining NSE,NF, GFAP were performed after BMSCs were transplanted into the vitreous and subretinal space in rat eyes for 30 days. Results 30 days after the transplantation, mophological and immunohistochemical examinations were implemented. New layer cells were founded. The morphology and alignment of these cells were similar to those of the retina of rat. Some of the cells expressed NSE, GFAP、 NF200. Conclusion BMSCs have the latent capacity to differentiate into retina-like structures.
