摘 要:目的探讨心脑舒通胶囊对脑缺血再灌注大鼠脑微循环障碍干预作用。方法采用线栓法制备大鼠局灶性脑缺血再灌注模型,24 h后处死取材,HE染色光镜下观察大鼠皮层区病理形态学变化和损伤状况;放射免疫法检测患侧皮层6-酮-前列腺素F1α(6-Keto-PGF1α)、血栓素B2(TXB2)水平;凝血酶法检测血浆纤维蛋白原(FIB)水平。结果缺血再灌注模型组患侧皮层有明显缺血性病理形态学改变,如神经细胞肿胀、血管内皮细胞肿胀、微血管管腔狭窄等;皮层6-Keto-PGF1α、TXB2水平异常升高(P<0.05,P<0.01);血浆FIB水平显著上升(P<0.01)。心脑舒通胶囊能明显减轻模型大鼠皮层病理形态学损伤;并可降低TXB2水平,降低血浆FIB水平(P<0.01,P<0.05)。结论心脑舒通胶囊可明显减轻缺血再灌注脑组织损伤,保护受损脑组织,其机理可能与调节血管活性物质、调整血液流变学病理状态、从而改善微循环障碍有关。Objective To investigate the intervene of Xinnaoshutong Capsules in brain microcirculation disorder in rats after cerebral ischemia and reperfusion. Methods The rat model of focal cerebral ischemia and reperfusion was established by using suture method. After 24 hours the rats were killed and the brain tissue was given HE staining. The pathomorphological changes and lesion in cortex were observed with the microscope, the levels of 6-Keto-prostaglandin F1α, (6-Keto-PGF1α) and thromboxane B2 (TXB2 ) in injured cortex were detected by applying radioimmunoassay, and the level of fibrinogen (FIB) was determined by using thrombin method. Results In the model group there were obvious ischemia pathomorphological changes in injured cortex, such as edema of neurons and vascular endothelial cells, and microvessel lumen stenosis. The levels of 6-Keto-PGF1α and TXB2 increased abnormally ( P 〈 0. 05, P 〈 0. 01 ), and plasma FIB level increased significantly ( P 〈 0. 01 ). Xinnaoshutong Capsules alleviated the pathomorphological lesion significantly, decreased the level of TXB2 and reduced plasma FIB level ( P 〈 0.01, P 〈 0.05 ). Conclusion Xinnaoshutong Capsules alleviates cerebral lesion induced by ischemia and reperfusion and protects injured brain tissue significantly. The mechanism may be related to that Xinnaoshutong Capsules can adjust vascular activated substance, regulate the pathological changes of hemorrheology, and relieve microcirculatory disturbance.
