摘 要:目的初步探讨携带bFGF-siRNA质粒表达载体的生物降解支架防治兔颈动脉再狭窄的作用。方法经股动脉入路建立球囊损伤兔颈动脉狭窄模型,实验动物分组后分别植入裸生物降解支架和携带bFGF-siRNA质粒表达载体的生物降解支架,分别以光镜和电镜观察狭窄段血管病理形态学变化,计算血管中膜和内膜厚度。结果裸支架组和siRNA支架组血管内膜厚度明显较对照组小,且siRNA支架组和裸支架组相比,血管内膜厚度明显减小,但三组之间中膜变化不明显。结论携带bFGF-siRNA质粒表达载体的生物降解支架能有效防治兔颈动脉再狭窄的发生,其具体分子机制尚需进一步的检测。Objective To investigate the preventative effect of biodegradable stent carrying with plasmid vector expressed bFGF siRNA on rabbit carotid restenosis. Methods Rabbit models with carotid stenosis were established by balloon injury via femoral artery approach. All the animals were divided into 3 groups and treated with control, biodegradable stent and biodegradable stent carrying with plasmid vector ex pressed bFGF siRNA, respectively. After the animals were sacrificed 4 weeks later, microscopy and electron microscopy were used to examine the thickness of the intima and media of the narrow vessel. Results The mean thickness of the intima of the bFGF siRNA stent group and naked stent group were significantly lower than that of the control group, and the difference between bFGF siRNA stent group and naked stent group was also significant, while the thickness of media among the three groups had no significance. Conclusion Biodegradable stent carrying with plasmid vector expressed bFGF siRNA can prevent carotid restenosis in rabbit, although the molecular mechanism needs to be clarified.
