摘 要:目的探讨补阳还五汤大鼠含药血清对家兔血小板PAF受体活性的影响。方法应用中药复方血清药理学研究方法,制备大鼠补阳还五汤含药血清,应用放射配基受体结合法观察大鼠含药血清对家兔血小板PAF受体活性的影响。结果补阳还五汤高、低剂量组,金纳多组和蒸馏水组对3H-PAF与家兔洗涤血小板PAF受体特异结合抑制率分别为51.4±15.7、44.3±11.3、52.2±12.2、34.3±20.3,各组间比较差异虽无统计学意义(P﹥0.05),但各用药组比蒸馏水组抑制作用分别提高了33.26%、22.57%、34.29%,其补阳还五汤高剂量组和金纳多组的抑制作用与蒸馏水组比较,统计量接近显著性水平(t=2.3082,t=2.6181,t24,0.05=2.8780)。结论补阳还五汤大鼠含药血清对家兔血小板PAF受体活性影响差异虽无统计学意义,但具有一定的抑制作用趋势,可能与其样本量较小或实验动物种属有关。Objective To explore the effects of the serum of rat fed with Buyang Huanwu Decoction (BYHWD)on the activity ofplatelet activating factor(PAF) receptor in rat. Methods According to Chinese medicine compound serum pharmacology method, the rat serums conraining BYHWD were produced. The effects of rat serum on activity of PAF receptor in rabbit were observed by using radioligand receptor binding assay. Results The specific binding inhibition ratio of high dose BYHWD group, low dose BYHWD group, Ginaton group and dis- tilled water group to 3H-PAF and rabbit washed platelet PAF receptor was 51.4± 15.7, 44.3± 11.3, 52.2±12.2, 34.3± 20.3 respectively. There was no statistical significance between groups(P 〉 0.05). But the inhibition effect of each medication group was respectively increased by 33. 26%, 22.57%, 34.29% compared with distilled water group.The inhibition effect of high dose BYHWD group and Ginaton group was compared with that of the distilled water group and the statistic was close to significant level (t = 2.3082, t = 2.6181, t24,0.06 = 2.8780). Conclusions There is no statistically significant difference in the effect of rat serum with BYHWD on activity of PAF receptor in rabbit.But it has a certain inhibition effect, which may be related to the relatively small sample size and the genera of the animal used for experiment.
