摘  要:采集发情期和间情期犬卵巢,回收大腔(直径大于1mm)卵泡和不可见卵泡的卵母细胞,在4种成熟液(TCM199+20%FCS,TCM199+20%FCS+10IU/mLFSH,TCM199+20%EDS和TCM199+20%EDS+10IU/mLFSH)中成熟后进行了受精试验。结果:(1)发情期大于2mm和1~2mm的大腔卵泡卵母细胞的体外成熟率(42.5%,32.5%)显著高于间情期卵母细胞(20.4%),但发情犬不可见卵泡卵母细胞的体外成熟率(28.9%)与间情期卵母细胞差异不显著;(2)在成熟液中添加促性腺激素,能有效地提高卵母细胞成熟率,不可见卵泡的卵母细胞对促性腺激素的依赖性(30.0%比7.8%)大于大腔卵泡的卵母细胞(42.5%比29.0%);(3)卵丘扩散与卵母细胞成熟存在相关性;(4)EDS可诱导卵丘扩散,但不能有效提高卵母细胞的成熟率;(5)发情期卵巢上的大腔卵泡和不可见卵泡的卵母细胞以及间情期卵巢不可见卵泡的卵母细胞体外培养后,与体外获能的精子进行体外受精,其卵裂率分别为16.0%(4/25),0%(0/40)和2.5%(1/40)。Dog ovaries were collected in estrous and diestrous period. The oocytes derived from large antral follicles (LAF>1 mm) and invisible follicles (IF) were recovered. They were cultured and matured in four kinds of mediums (TCM199+10 IU/mL FSH+20%FCS; TCM199+20%EDS+10 IU/mL FSH; TCM199+20%EDS and TCM199+20%FCS) for up to 72 h and then fertilized in vitro. The results were as follows: ①In vitro maturation rate of LAF oocytes of the estrous ovaries (42.5%, 32.5%) was significantly higher than that of the diestrous ovaries (20.4%). ②Gonadotropins effectively improved in vitro maturation rate of dog oocytes, but the oocytes recovered from IF were more sensitive to gonadotropins than those from LAF. But there were no differences in the maturation rates of IF oocytes between estrous and diestrous. ③The expantion of cumulus was related to the maturation of oocytes. ④EDS could induce the expantion of cumulus cells, but did not improve effectively the maturation rate of oocytes. ⑤When three kinds of oocytes (oocytes from LAF, oocytes from IF in both estrous and diestrous ovaries) cultured in TCM199+20%FCS+10 IU/mL FSH for up to 72 h were fertilizeed in vitro with capacited sperms the oocyte cleavage rate was 16 0% (4/25), 0%(0/40) and 2.5%(1/40), respectively.
