摘 要:目的研究白术挥发油的化学成分及对S180荷瘤小鼠的抗肿瘤作用。方法采用水蒸气蒸馏法提取得到白术挥发油,常规硅胶柱层析将白术挥发油按极性分成极性大的部分(zbo-2)和极性小的部分(zbo-1),并分离纯化白术挥发油的化学成分,用核磁共振和X-射线衍射法鉴定其结构。并以肿瘤生长抑制率为指标观察白术内酯Ⅰ、白术内酯Ⅲ、zbo-2、zbo-1对S180荷瘤小鼠的抗肿瘤作用。结果共从白术挥发油中分离得到3个化合物,分别为白术内酯Ⅰ、白术内酯Ⅱ、白术内酯Ⅲ。白术内酯Ⅰ、白术内酯Ⅲ、zbo-2、zbo-1小剂量和zbo-1大剂量对S180荷瘤小鼠的抗肿瘤作用均较弱,其中zbo-1大剂量(200 mg/kg)抗肿瘤作用最强,抑瘤率为24.74%。结论白术内酯Ⅱ为第1次从白术挥发油中分离得到;白术挥发油的抗肿瘤有效成分可能存在于白术挥发油中极性小的部位。Objective To study the chemical compositions of volatile oil from Rhizorna Atractylodis Macrocephalae and its anti-tumor effect in mice bearing tumor S180. Methods The volatile oil from Rhizoma Atractylodis Macrocephalcte was extracted by steam distillation, which was then divided into two parts according to its polarity, i.e. part of high polarity (zbo-2) and part of low polarity ( zbo-1 ), by using general silicagel column chromatography. The chemical compositions of the volatile oil were separated and purified at the same time, and its structure was determined by nuclear magnetic resonance (NMR) and X-ray diffraction method. The anti-tumor effects of atractylenolide Ⅰ, atractylenolide Ⅲ, zbo-2 and zbo-1 in mice bearing tumor S180 were observed taken tumor growth inhibitory rate as the index. Results There were 3 compounds separated from the volatile oil from Rhizoma Atractylodis Macrocephalae, including atractylenolide I, atractylenolide II and atractylenolide III. Atractylenolide I, atractylenolide III, zbo-2 and zbo-1 in low dose and high dose had a weak anti-tumor effect, but zbo-1 in high dose (200 mg/kg) had a stronger anti-tumor effect (tumor growth inhibitory rate was 27.74% ). Conclusion Atractylenolide II was separated for the first time from the volatile oil from Rhizoma Atractylodis Macrocephalae. The anti-tumor active principle may be in the part of low polarity ( zbo-1 ) of the volatile oil from Rhizoma Atractylodis Macrocephalae.
