摘 要:目的观察益气补肾中药脑神康胶囊对体外培养大鼠海马神经元缺氧复氧损伤的保护作用。方法原代培养大鼠海马神经元,随机分为对照组,损伤组,脑神康胶囊高、中、低浓度组。对照组正常培养,损伤组及脑神康胶囊各浓度组建立缺氧复氧损伤模型,其中脑神康胶囊各浓度组于复氧时换以不同浓度(5%、10%、20%)含药血清的培养液,检测各组细胞超氧化物歧化酶(SOD)、乳酸脱氢酶(LDH)活力及丙二醛(MDA)含量,测定细胞存活率及凋亡率,RT-PCR法检测bcl-2 mRNA和bax mRNA的表达及两者比值。结果与对照组比较,损伤组SOD活性及细胞存活率均明显下降(P<0.01),MDA含量、LDH活力及细胞凋亡率均显著升高(P<0.01),bcl-2 mRNA表达下降,bax mRNA表达升高,bcl-2/bax降低(均P<0.01);脑神康胶囊各浓度组均较损伤组SOD活性及细胞存活率均升高(P<0.01),MDA含量及细胞凋亡率均降低(P<0.01),LDH活力及bax mRNA表达降低(P<0.01,P<0.05),bcl-2 mRNA表达升高,bcl-2/bax增加(均P<0.01),且呈明显的剂量依赖性。结论脑神康胶囊对海马神经元缺氧复氧损伤有明显的保护作用,其作用机制可能与其提高神经元抗氧化能力、调节凋亡相关蛋白、减少凋亡密切相关。Objective To explore the protective effects of Naoshenkang-traditional Chinese medicine with function of invigorating Qi and tonifying kidney against neuronal anoxia-reoxygenation injury and to investigate its mechanisms. Methods The primarily cultured neonate rat hippocampal neurons were randomly divided into normal, anoxia-reoxygenation and large, middle, and small dosage of Chinese Med- icine-protective groups. The Chinese Medicine-protective groups were treated with different concentrations of Naoshenkang containing serum before reoxygenation. The metabolic rate of MTF, the content of malondialdehyde (MDA) and the activities of superoxide dismutase (SOD), lactate dehydrogenase (LDH) were measured by assay^kits. The apoptosis rates of hippocampal neurons were tesed by flow eytometry. RT:PCR was used to evaluate the mRNA expressiogs of bcl-2 and bax genes. Results The activity of SOD, survival rate of cell, the ratio of bcl-2/bax and the expression of bcl-2 mRNA in anoxia-reoxygenation group were significantly lower ( all P 〈 0. 01 ), but the levels of MDA and LDH, the apoptosis rate and the expression of bax mRNA were higher ( all P 〈 0. 01 ) than those in normal group. The activity of SOD, the survival rate of cell, the expression of bcl-2 mRNA and the ratio of bcl-2/bax in all Chinese Medicine-protective groups were significantly higher ( all P 〈 0. 01 ) , but the levels of MDA and LDH, the apoptosis rate and the expression of bax mRNA were lower ( P 〈 0. 01, P 〈 0. 05 ) than those in anoxia-reoxygenation group and demonstrated obvious dosage-effect relationship. Conclusions Naoshenkang capsule has a protective effect on hippocampal neurons against anoxia-reoxygenation injury, and its mechanism maybethat the Chinese Medicine can increase the antioxidative activities of neurons, regulate associated protein of apoptosis and inhibit the apoptosis.
