绞股蓝颗粒对糖尿病肾病大鼠肾脏血管紧张素及内皮素表达的影响

摘　　要：目的:观察绞股蓝颗粒对糖尿病肾病模型大鼠血管紧张素及内皮素表达的影响,以了解其治疗糖尿病肾病的作用机制。方法:利用单侧肾切除加STZ注射法改良复制糖尿病肾病大鼠模型,实验分为正常组、模型组、治疗组(绞股蓝颗粒高、中、低剂量组及缬沙坦对照组),观察糖尿病肾病模型大鼠肾组织AngⅡ,ET-1含量变化,AT1RmRNA及ETARmR-NA的表达。结果:治疗组肾组织AngⅡ,ET-1含量显著减少,AT1RmRNA及ETARmRNA明显下调,与模型对照组比较,差异有统计学意义(P<0.05或P<0.01)。结论:绞股蓝颗粒可用于治疗早期糖尿病肾病,其机制可能与其调节RAS系统及内皮素系统紊乱有关。Objective： Observing the effect of on the renal angiotension and endothelin＇s expression in diabetic nephropathy, to find out the possible mechanism of the therapeutical effect of gynostemma pentaphylla powder to diabetic nephropathy. Methods： The experimental rats were divided into six groups：normal group, model group and treatment groups（ high,middle, low dose groups of gynostemma pentaphylla powder and the Valsartan group）. The content of the angiotension II and endothelin - 1 in kidney, the expression of mRNA of angiotensinogen receptor 1 and endothelin receptor A in kidney were observed. Results： The content of the angiotension II and endothelin- 1 in kidney, the expression of mRNA of the angiotensinogen receptor 1 and endothelin receptor A in kidney in treatment groups was lower than the model group, the difference were significant（ P 〈 0.05 or P 〈 0.01 ）. Conclusion： Gynostemma pentaphylla powder can be used to treat early diabetic nephropathy, and the mechanism maybe is regulating the disorder of the renin - angiotension system and the endothelin system.
