摘 要:采用经过优化后的DNA-AFLP分析体系,分析了春兰原变种与线叶春兰花基因组上的差异.9对引物组合在100~800 bp之间共扩增出389条带.其中共有条带318条,春兰原变种有11条特异性条带,而线叶春兰有60条特异性条带.线叶春兰特异带的48.3%集中500~800 bp,51.7%集中在100~500 bp,春兰原变种特异带全部集中500~800 bp.通过对不同引物分析发现引物对(E-AGC/M-CAT)对春兰原变种以及线叶春兰的特异性片段选择性较好.两种兰花花基因组上的差异可能与两种兰花花形态,以及香味差异有密切关系.The flowers of Cymbidium goeringii var. goeringii and the flowers of Cymbidium goeringii var. serratum (Schitr.)Y. S. Wu et S. C. Chen were analyzed by DNA-AFLP after the system of AFLP was optimized. 389 bands ranging with 100-800 bp were amplified with 9 pairs of primers. 318 bands were owned by both. There were 11 different bands in the var. goeringii, and 60 specific bands in the var. serratum. 48.3% of the specific bands in the var. serratum concentrated on the range of 500-800 bp and 51.7% of them on the range of 100-500 bp. All of the specific bands in the var. goeringii concentrated upon the range of 500~800 bp. The primers (E-AGC/M-CAT)could get more different bands for both the var. goeringii and the var. serratum by selective amplification. The difference of genome could have close relation with the difference of morphology and aroma between two variations.
