摘  要:[目的]对从水稻幼苗中分离的2个3型金属硫蛋白基因OsMT-I-3a和OsMT-I-3b的结构进行了分析,并对其表达模式进行了检测。[方法]利用Blast搜索水稻数据库,设计引物并利用RT-PCR的方法克隆OsMT-I-3基因cDNA全长。通过多重比对等方法对其基因和蛋白结构进行了分析。利用Northern杂交对这两个基因的组织表达模式和环境应答模式进行了检测。[结果]OsMT-I-3a和OsMT-I-3b分别编码62和65个氨基酸的多肽,具有典型的3型金属硫蛋白的半胱氨酸分布模式。OsMT-I-3a基因在叶片中没有检测到表达,OsMT-I-3b在叶片中高表达,且各种非生物胁迫因素会影响两种基因的表达模式。[结论]Ⅰ类3型2个水稻MT基因可能参与水稻多种环境胁迫的信号通路,在水稻抗非生物胁迫过程中起作用。XU Yu-feng et al ( Laboratory of Molecular Biology and Protein Science, Department of Biological Sciences and Biotechnology, Tsinghua University, Beijing 100084) Abstract [ Objective] The research aimed to analyze the structure of two type-3 metallothionein genes OsMT-I-3a and OsMT-I-3b isolated from rice seedlings and detect their expression patterns. [ Method] Using Blast, rice database was searched to design the primers. Full-length eDNA of OsMT-I-3a and OsMT-I-3b was cloned by using RT-PCR. The structure of genes and proteins were analyzed by using sequence comparison and other methods. The tissue expression pattern and the stress response models of two genes were tested by using Northern blot. [ Result ] OsMT-I-3a and OsMT-I-3b, encoded by 62 and 65 encoding peptide amino acids respectively, had typical type-3 metallothionein cysteine distribution patterns. The expression of OsMT-I-3a was not detected in the leaves of rice, but OsMT-I-3b gene was obviously expressed in the leaves. Moreover, various abiotic stress factors affected the expression of OsMT-1-3a and OsMT-1-3b. [ Conclusion] I -class type-3 rice MT genes may be involved in a wide range of environmental stress signaling pathway and play an important role in antiabiotic stress of rice.
