摘 要:目的研究强脉冲光(IPL)对皮肤成纤维细胞TGF—β1表达的影响,探索IPL影响皮肤胶原合成的调控机制。方法将体外分离培养的人包皮原代成纤维细胞,分成照射组和对照组,照射组以波长590~1200nm、能量密度为29J/cm^2、脉宽15ms、延时5ms的IPL对成纤维细胞进行照射。两组细胞均采用无血清DMEM分别培养1、12、24、48h,然后荧光定量PCR法分别测定4个时间点细胞TGF-β1的表达情况。结果培养12、24、48h后,TGF—β1的表达在照射组成纤维细胞中明显增高,与对照组比较,差异有统计学意义(P〈0.01)。结论IPL能促进成纤维细胞合成TGF—β1的活性。Objective To investigate the effects of intense pulsed light (IPL) on the expression of TGF-β1 in human fibroblasts. Methods The primary foreskin fibroblasts cultured in vitro were divided into the control group and the experimental group. The fibroblasts in the experimental group were irradiated by the IPL with 590 - 1200 nm wave length, 29 J/cm^2 energy density and 15 ms pulse width. The fibroblasts from 2 groups were cultured with non - serum DMEM for 1, 12, 24 and 48 hours after the irradiation, the expression of TGF-β1 in two groups at different time was measured respectively by fluorescent PCR (FQ-PCR) technique. Results The expression of TGF-β1 of fibroblasts in the experimental was increased and significandy higher than those in the control group at 12, 24 and 48 hours after IPL irradiation. Conclusion IPL irradiation can increase the synthesis of TGF-β1 in cultured primary fibroblasts.
