摘 要:目的了解四川资阳地区健康猪携带猪链球菌状况及其主要毒力基因分布和药物敏感性,分析这些菌株与病人分离株的同源性,为疫情预测和制定防治措施提供依据。方法从屠宰场采集健康猪的咽拭子和扁桃体标本分离培养获得分离株,用API 20 Strep进行生化鉴定、猪链球菌诊断血清进行血清分型;聚合酶链反应(PCR)检测猪链球菌种特异性基因(16S rDNA)和相关毒力基因;K-B纸片法进行药物敏感性分析;并用脉冲场凝胶电泳(PFGE)分析菌株的同源性。结果从健康猪分离到了12株猪链球菌,均为血清2型;菌株均具有cps2J、mrp、sly、ef和gapdh毒力基因;所有菌株抗药性一致;PFGE分析结果显示均为SmaI 001型,与同期病人分离株带型一致。结论四川资阳地区健康猪携带的2型猪链球菌是高致病性菌株,与同期病人分离株具有一致的生物学特征,是引起猪或人发病的重要传染源;猪带菌率存在季节差异。To investigate the carrying status of Streptococcus suis, the distribution of virulence genes and the antibiotics susceptibility of the isolates in healthy pigs in Ziyang district, Sichuan province, and to analyze the homology between isolates from pigs and those isolated from human patients , so as to provide a basis for outbreak prediction and preventive measures, all strains of Streptococcus were isolated from samples of nasopharyngeal swabs and tonsils collected from healthy pigs , bioehemically identified with API 20 Strep kit and serologically typed with specific diagnostic sera for S. suis. Polymerase chain reaction (PCR) was used to detect the presence of their specific gene 16S rDNA and the virulence-related genes. The antibiotics susceptibility of the isolates was determined by K-B paper disc method and the the homology between isolates from pigs and human patients was analyzed by pulse-field gel electrophoresis (PFGE). It was found that 12 strains of S. suis were isolated from healthy pigs and all the isolates were proved serologically to be type 2. All the isolates carried virulence genes ,such as cps2J, mrp, sly, ef and gapdh and had the same antibiotics susceptibility spectrum. As demonstrated by PFGE pattern, the isolates from pigs were indistinguishable with those from human patients during the same time. From these observation, it is concluded that the strains of S. suis isolated from hea]tby pigs in Ziyang district of Sichuan province are proved to be highly pathogenic and highly homologous to those strains isolated from human patients. However, the occurrence of S.suis type 2 in healthy pigs shows seasonal disparity.
