摘　　要：为了探讨自体角朊细胞体外对人-猪异种混合血管内皮细胞-淋巴细胞反应(MELR)的影响及作用机制,我们从mR-NA;蛋白水平观察淋巴细胞的增殖;细胞因子的分泌及胞内细胞因子的变化,并用IL-10单抗进行封阻,观察其对角朊细胞诱导免疫抑制效应的影响。结果发现在MELR中引入自体角朊细胞后,细胞因子分泌格局呈现由IFN-γ主导转变为由IL-10主导的动力学变化;IL-10单抗能有效地阻断角朊细胞所诱导的免疫抑制效应。表明自体角朊细胞可诱导一群分泌IL-10的调节性T细胞,从而抑制Th1亚群所介导的细胞免疫及异种移植物排斥反应。To investigate the effect of keratinocytes （KCs） on lymphoproliferation to xenogeneic antigens, a mixed swine mixed endothelial-lymphocyte culture （MELC） system was established, and the effect of autologous KCs on the MELC were examined by using 3H-TdR incorporation, RT-PCR and ELISA to determine the cell proliferation intensity and secretion levels of Th1/Th2-related cytokines. In this study, it was shown that the human KCs that were superimposed on MELC could induce strong suppression on the responses of autologous lymphocytes to swine endothelial cell line （PIEC）. At the same times, the Thl-related cytokine pattern, IFN-γ^＋ IL-4^- IL-10^-, was completely shifted to a condition dominant by the IFN- γ^-IL-4^-IL-10^＋ pattern, when autologous, instead of allogeneic KCs were used as inducers. The IL-10 blocking test could reverse the suppression effect of auto-KCs on MELC. This implicates that the auto-KCs might induce transplantation tolerance by interfering the Thl-mediated xeno-responses via activating an IL-10-secreting T subset with down regulatory activity. This effect might provide a new strategy to induce tolerance in xeno-transplantation.
