摘 要:目的观察重组人血小板型磷脂酶A2(PLA2)对革兰阳性球菌的抗生素后效应(PAE),探讨血小板型PLA2的杀菌机制。方法采用微量稀释法测定重组人血小板型PLA2对3种革兰阳性球菌(金黄色葡萄球菌、表皮葡萄球菌、粪肠球菌)的最低抑菌浓度(MIC)。在不同药物浓度作用下,采用平板倾注法对细菌行菌落计数单位(CFU)计数,计算PAE。结果血小板型PLA2对金黄色葡萄球菌、表皮葡萄球菌、粪肠球菌的MIC分别为60、80、120 ng/ml,血小板型PLA2对这3种细菌均有明显的PAE,对金葡菌的PAE最长,表皮葡萄球菌次之,粪肠球菌的最短。结论血小板型PLA对革兰阳性细菌有明显的PAE。Objective To study the postantibiotic effect(PAE) of recombinant human platelet phospholipase A2(PLA2) to the three gram-positive(G+) bacteria(Staphylococcus aureus,Staphylococcus epidermidis,Enterococcus facecalis)and exploring bactericidal mechanism of the PLA2.Methods The minimal inhibitory concentration(MIC) of the PLA2 to three G+ bateria was determined by microdilution method.At different concentration,The number of colonies was counted by pourplate,then calculated PAE.Results MIC of the thrombocyte PLA2 to Staphylococcus aureus, Staphylococcus epidermidis, Enterococcus facecalis were seperaly 60, 80, 120 ng/ml. Thrombocyte PLA2 presented obviously PAE to the three G^+ bacteria. The PEA of Staphylococcus aureus was the longest, followed by Staphylococcus epidermidis, Enterococcus faeecalis was the least. Conclusion Recombinant human thrombocyte PLA2 presentes obviously PAE to the G^+ bacteria.
