金边瑞香活性成分瑞香黄烷-Ⅰ,瑞香素对SMMC-7721和MCF-7细胞作用的研究

摘　　要：目的研究金边瑞香活性成分瑞香黄烷-Ⅰ(E002)、瑞香素(E003)抗肿瘤活性,为进一步研究抗肿瘤机制提供依据。方法用噻唑蓝(MTT)法检测不同浓度E002、E003对SMMC-7721和MCF-7细胞的抑制率。结果①E002、E003浓度为250~1.95μg/m l时,作用SMMC-7721细胞,两者的抑制率为:(86.9±2.2)%^(50.8±0.6)%。②E002浓度为250~62.5μg/m l时,作用MCF-7细胞,其抑制率为:(72.3±2.1)%^(52.48±1.65)%。③E002对SMMC-7721和MCF-7细胞的IC50值分别为:(1.96±0.03)和(22.89±3.04)μg/m l;E003对SMMC-7721细胞的IC50值为:(1.23±0.11)μg/m。l结论金边瑞香活性成分瑞香黄烷-Ⅰ对SMMC-7721、MCF-7细胞,瑞香素对SMMC-7721细胞的增殖均有显著抑制作用,且呈现良好的剂量依赖性。Objective To study the antitumor activity of Daphne odor var. marginata extracts - E002 （ daphnodorin I） and E003 （daphnetin） in vitro. Methods MCF -7 and SMMC -7721 cells were treated respectively with E002 and E003 at different concentrations, the growth inhibitory effects of E002, E003 were assessed using the MTT assay . Results （ 1 ） Both inhibition rate （%）were from 86.9 ± 2.2 to 50.8 ± 0.6,when SMMC -7721 cells were treated with E002, E003 in the concentration range of 250 - 1.95 （μg/ml）. （2） Inhibition rate （ % ） were from 72.3 ±2.1 to 52.48± 1.65, when MCF - 7 cells were treated with E002 in the concentration range of 250 -62.5 （μg/ml）. （3）IC50 value E002 on SMMC -7721 and MCF -7 cells （μg / ml） were 1.96 ± 0.03 and 22.89 ±3.04 ;IC50 value E003 on SMMC - 7721 cells （ μg / ml） was ： 1.23±0.11. Conclusion - Daphnodorin I has significant inhibitory effect on MCF- 7 and SMMC -7721 cells in a dose -dependent in vitro. Daphnetin has significant inhibitory effect on SMMC -7721 cell in a dose -dependent in vitro.
