摘  要:目的探讨健脾活血方对2型糖尿病胰岛素抵抗(Insulin Resistance,IR)大鼠葡萄糖转运蛋白-4(Glucose Trans-porter-4,GLUT-4)mRNA表达的的改善作用及机理。方法取Wistar大鼠60只,采用链脲佐菌素(Streptozotocin,STZ)加高脂饮食造成2型糖尿病IR大鼠模型,设空白组、模型组、健脾活血方低剂量组、健脾活血方高剂量组和罗格列酮组,以RT-PCR法检测大鼠骨骼肌GLUT-4 mRNA的表达。结果健脾活血方高剂量组大鼠骨骼肌GLUT-4 mRNA的表达与罗格列酮组相当,与模型组比较有显著性差异(P<0.01)。结论健脾活血方能够上调大鼠骨骼肌GLUT-4 mRNA的表达,改善IR。Objective To explore the effect and mechanisms of Trengthening Spleen and Activating Blood Prescription on expressions of GLUT - 4 gene in the skeletal muscle tissue of Type 2 Diabetes Mellitus rats. Methods 60 Wistar rats were randomized into five groups : normal control group, model group, Strengthening Spleen and Activating Blood Prescription low - dose group and high - dose group, Rosiglitazone group. The model of Type 2 Diabetes Mellitus IR was established by adopting STZ and high - fat diet, GLUT - 4 gene expression in skeletal muscle tissue was detected by RT - PCK. Results The expression of GLUT - 4 gene in Strengthening Spleen and Activating Blood Prescription high -dose group was the same with Rosiglitazone group's, and there was significant difference compared to model group (P 〈 0.01 ). Conclusion Strengthening Spleen and Activating Blood Prescription can up - regulate the expression of GLUT - 4 gene in Type 2 Diabetic rats, and improve insulin resistance.
