摘  要:目的建立龙胆最佳提取工艺,为龙胆的种植和质量研究奠定基础。方法采用L9(34)正交表安排实验,确定最佳提取条件。色谱柱为Shimadzu C18(3.0 mm×75 mm,1.7μm);流动相为乙腈-0.4%磷酸(V∶V=5.5∶94.5),流速0.80ml/min;检测波长235,270 nm;柱温为40℃。结果影响因素最大的是提取溶剂,其次是提取方法,最小的是提取时间。结论各因素较优的水平组合为A1B1C1,即甲醇超声提取30 min。Objective To establish best extraction process of Gentiana scabra Bge. by UPLC so as to provide base for the plant and research of Gentiana scabra Bge. Methods By L9 ( 3^4 ) orthogonal design , the optimal condition was confirmed as follows : Shimadzu C18(3.0 mm ×75 mm,1.7μm), Acetonitrile -0.1% Phosphorylation(V: V = 5.5: 94.5) as nwbile phase the flow rate was 0.8 ml/min, detection wavelength was 235nm and 270nm and the columne temperature was 40℃. Results The extraction solution was the most effective factor , the second was the method, and the last was the extraction time. Conclusion The best extraction process is A1 B1C1 : super sonic extraction for 30 min in methanol.
