摘 要:目的比较决明子总蒽醌的水提和醇提工艺,确定提取决明子总蒽醌的最佳工艺。方法采用正交实验法,分别在水提工艺中考察了浸泡时间、提取时间、提取次数和固液比,在醇提工艺中考察了乙醇浓度、提取温度、提取时间和固液比对决明子总蒽醌提取率的影响。用紫外-可见分光光度法测定总蒽醌的含量。结果总蒽醌的提取量方面醇提效果明显优于水提。在醇提的考察因素中,乙醇浓度、提取时间和固液比对总蒽醌的提取有显著影响(P<0.05)。结论决明子总蒽醌提取的最佳工艺为8倍用量的50%乙醇在80℃下提取2h,提取次数2次。Objective To compare the effect of water extracting and alcohol extracting processes on anthraquinone content in Semen cassiae and to optimize the extraction of total anthraquinone of Semen cassiae. Methods Choosing water as the solvent, the permeating time, the extracting time, the extracting frequencies and the amount of solvent were determined by orthogonal test. With different concentrations of alcohol as the solvent, the extracting temperature, the extracting time and the amount of solvent were determined by orthogonal test. The content of total anthraquinone was determined by UV - Vis Spectrophotometry. Results Alcohol was a better solvent to extract anthraquinone than water. Using alcohol as the solvent, the concentration of alcohol, the extracting time and the amount of solvent were significant factors (P 〈 0. 05 ). Conclusion The optimum extraction process is as follows: 80℃ ,reflux extraction with 8 -fold 50% alcohol for 2 times ,2h per time.
