灯盏花素改善肿瘤化疗药物对小鼠lewis肺癌的治疗指数

摘　　要：目的:观察灯盏花素对肿瘤化疗药物治疗效果的影响。方法:MTT法检测灯盏花素体外对阿霉素、环磷酰胺或氟脲嘧啶诱导脾脏细胞凋亡的保护作用;将lewis肺癌细胞接种于C57BL/6小鼠皮下和肺,采用灯盏花素联合阿霉素、环磷酰胺或氟脲嘧啶治疗小鼠,观察肿瘤大小、外周血细胞数、骨髓细胞数、脾细胞凋亡、及小鼠存活时间。结果:灯盏花素体外对阿霉素、环磷酰胺或氟脲嘧啶诱导的脾脏细胞凋亡有保护作用。在体研究,治疗组均有比未治疗组不同比例的肿瘤生长减慢,单独化疗导致骨髓细胞和外周血细胞数减少、脾细胞凋亡增加。实验结束时,与单一药物治疗相比,灯盏花素联合化疗均能显著延长小鼠存活时间。单独化疗虽比未治疗组小鼠存活时间稍长,但无统计学差异。结论:单独化疗对伴随肿瘤转移的荷瘤小鼠生命延长无实际意义,灯盏花素能改善肿瘤化疗药物的治疗指数。Objective： To observe the effect of breviscapine on therapeutic efficacy of chemotherapeutic agents for tumors. Methods： The protective effect of breviscapine on spleen cells against doxorubicin, cyclnphosphamide and fluorouraeil induced apoprosis was examined by MTT assay in vitro. LLC cells were injected subcutaneously or intravenously into C57BL 6 mice. The mice were treated with the combination of breviscapine with doxorubicin, cyclophosphamide or fluorouracil. The therapeutic efficacy was evaluated by the tumor size, the peripheral blood cells, the hone marrow cells, spleen cell apoptosis and mouse＇ s life span. Results： Breviscapine could protect spleen cells against doxorubicin, cyclophosphamide and fluorouracil induced apoplosls in vitro. In vivo, the tumor size in the treated mice was significantly smaller than that of the untreated mice. The chemotherapy alone resulted in the reduction in the peripheral blood cells and bone marrow cells, and the increase in spleen cell apoplosis. At the end of experiment , the combination treated mice with hreviscapine and other chemotherapeutic agents had the longer life span than that of the single agent treated mice. The life span slightly prolonged in the mice treated with chemotherapy alone, but the difference was not statistically significant, Conclusion： The chemotherapy alone is not significant to life span in tumor bcaring mice with metastasis, breviscapine can improve therapeutic index of chemotherapeutic agents for tumor.
